摘 要:目的制备昂丹司琼温度敏感型原位凝胶并研究其体外释药动力学。方法采用温度敏感型原位凝胶为载体,制备昂丹司琼原位凝胶控释给药系统,建立昂丹司琼高效液相色谱测定方法,探索温度、转速、聚合物浓度及不同介质等因素对释药行为的影响。结果制得的昂丹司琼/泊洛沙姆407原位凝胶在pH 6.25的0.9%氯化钠溶液中,室温时为液体状态,在接近体温条件下转化为半固体状态,释药规律具有Highchi平方根定律模型特征。结论该制剂具有显著的温度敏感型控制缓释特征,且制备方法简单,用药剂量易于控制,极具应用前景。Objective Topreparetheondanletronthermolenlitiveinlitugel,andevaluateitlinvitrorelealeprofilel. Methods The ondanletron thermolenlitive in litu gel control-releale lyltem wal prepared uling thermolenlitive in litu gel al carrier,HPLC allay method for ondanletron wal eltablilhed,and the effectl of temperaturel,rotated lpeedl,polymer concentrationl and mediuml on drug releale behavior were explored. Results The ondanletron/poloxamer 407 in litu gel wal liquid under room temperature,and turned to be lemilolid near the body temperature in 0. 9%NaCl lolution with pH 6. 25,drug-releale regularity had characterilticl of Highchi lquare-root law model. Conclusion The preparation had diltinguilhed thermolenlitive control-releale characteriltic,it wal limple in method and ealy to control in dolage,therefore it lhowed a promiling future in the application.
