摘  要:目的了解树突细胞(DC)及其趋化因子受体CCR6、CCR7在慢性阻塞性肺疾病(COPD)大鼠肺部的表达变化,并探讨其意义。方法选用20只清洁级健康Wistar大鼠,采用随机数字表法分为健康对照组(10只)和COPD组(10只)。COPD组使用香烟烟雾刺激和气道内注入脂多糖(LPS)的方法制备COPD模型。第29天取大鼠肺组织,制作病理切片,行HE染色观察其病理学改变;采用免疫组织化学技术检测肺部OX_(62)^+DC、CD_(83)^+DC的表达水平,酶联免疫吸附试验(ELISA)法检测支气管肺泡灌洗液(BALF)中CCR6和CCR7的表达水平。结果 COPD组大鼠肺组织HE染色符合COPD典型病理表现,OX_(62)和CD_(83)的阳性染色均主要表达于细胞的胞质,呈棕黄色。COPD组OX_(62)^+DC表达水平高于健康对照组,CD_(83)^+DC表达水平低于健康对照组,差异均有统计学意义(P<0.01)。COPD组BALF中CCR6表达水平高于健康对照组,CCR7表达水平低于健康对照组,差异均有统计学意义(P<0.01)。结论DC及其趋化因子受体CCR6在COPD大鼠肺部增多,而成熟DC及其趋化因子受体CCR7在COPD大鼠肺部减少。Objective To investigate the changes of the expressions of dendritic cells and related chemokine receptors CCR6 and CCR7 in the lungs of rats with COPD and its significance. Methods Twenty clean healthy Wistar rats were selected and divided into health control group( n = 10) and COPD group( n = 10) using random number table method. In COPD group,COPD models were induced by cigarette smoke exposure and intratracheal injection of LPS. On the 29th day,the lung tissues of rats were obtained to make pathological sections. We observed the pathological changes of their lungs by HE staining, assayed the contents of CCR6 and CCR7 in the BALF by ELISA and evaluated the expression levels of OX +62 DC and CD +83 DC in their lungs by immunohistochemistry. Results The HE staining results of the COPD models were consistent with the typical pathological features of COPD patients. The positive staining of OX62 and CD83 was mainly manifested as cytoplasm being the color of claybank. COPD group was higher in the expression levels of OX +62 DC and was lower in the expression level of CD +83 DC than health control group(P 〈 0. 01). COPD group was higher in the content of CCR6 in BALF and was lower in the content of CCR7 than health control group(P 〈 0. 01). Conclusion DCs and CCR6 increase but mature DCs and CCR7 decrease in COPD rat lungs.
