摘  要:目的:探讨夏枯草提取物对急性心力衰竭(AHF)模型大鼠心肌保护作用及基质细胞衍生因子-1(SDF-1)表达的影响,探讨其治疗AHF的可能作用机制。方法:采用Wistar大鼠建立AHF模型,随机分为4组,AHF模型组(AHF组),给予等体积生理盐水ig;夏枯草低剂量组,按照夏枯草提取物0.5 g·kg-1剂量ig;夏枯草中剂量组,按照夏枯草提取物1.0 g·kg-1剂量ig;夏枯草低剂量组,按照夏枯草提取物2.0 g·kg-1剂量ig;另设假手术组(Sham组),给予等体积生理盐水ig。6周后进行血流动力学、超声心动图测量,并分别采用流式细胞术,苏木素-伊红(HE)染色测量CD34+细胞阳性率,微血管密度(MVD)。进一步采用蛋白质免疫印迹(Western blot)和实时荧光定量-聚合酶链式反应(q PCR)分析检测SDF-1蛋白和SDF-1mRNA表达的变化。结果:夏枯草提取物可有效改善AHF大鼠左心室舒张功能,减轻心室壁厚度,促进CD34+细胞的动员和新生血管的形成,与AHF组比较,差异有统计学意义(P<0.05,P<0.01)。夏枯草提取物可使SDF-1蛋白和SDF-1 mRNA的表达显著增加,与AHF组比较,差异有统计学意义(P<0.05,P<0.01)。结论:夏枯草提取物对衰竭心脏有明显的保护作用,其机制可能与促进新生血管的形成,升高SDF-1表达水平和动员CD34+细胞释放至外周血有关。Objective: To investigate the protective effect of Prunellae Spica extract on the myocardium and its effect on the expression of stromal cell derived factor-1( SDF-1) in acute heart failure( AHF) rat models and discuss its possible mechanism. Method: Wistar rats were used to establish AHF models and randomly divided into four groups, such as AHF model group( AHF group) with equal volume of normal saline by intragastric administration; Prunellae Spica low-dose group,0. 5 g·kg- 1Prunellae Spica extract by intragastric administration; Prunellae Spica middle-dose group, 1. 0 g ·kg- 1Prunellae Spica extract by intragastric administration; and Prunellae Spica high-dose group, 2. 0 g ·kg- 1Prunellae Spica extract by intragastric administration. Another sham operation group( Sham group) was set to receive equal volume of normal saline by intragastric administration. After 6 weeks,hemorheology examination and ultrasonic cardiogram were taken. The positive rate of CD34+cells and microvessel density( MVD) were measured by flow cytometry and HE staining respectively. Western blot and q PCR were further used to detect the protein and mRNA expressions of SDF-1.Result: Prunellae Spica extract can effectively improve left ventricular diastolic function,reduce ventricular wall thickness and promote formation of CD34+cell mobilization and angiogenesis in AHF rat models,with statistically significant difference as compared with AHF group( P 〈 0. 05, P 〈 0. 01). Prunellae Spica extract can significantly increase the expressions of SDF-1 protein and SDF-1mRNA,with statistically significant difference as compared with AHF group( P 〈 0. 05,P 〈 0. 01). Conclusion: Prunellae Spica extract has obvious protective effect on heart failure,and the mechanism may be related to promoting angiogenesis,increasing SDF-1 expression levels and mobilizing the release of CD34+cells to peripheral blood.
