新疆维吾尔族哮喘儿童IL-13基因Arg130Gln多态性及其与血清IL-13水平的关系

摘　　要：目的探讨新疆维吾尔族哮喘儿童白细胞介素-13（IL-13）基因Arg130Gln多态性及其与血清IL-13水平的相关性。方法用聚合酶链反应（PCR）及直接基因测序法对38例维吾尔族哮喘儿童及30例健康儿童行IL-13基因Arg130Gln多态性分析,采用酶联免疫吸附（ELISA）法对哮喘儿童及健康儿童进行血清IL-13水平的测定,并对两组儿童血清IL-13水平进行分析。结果 （1）哮喘组与对照组IL-13基因Arg130Gln位点基因型及等位基因频率分布差异均无统计学意义（基因型：χ^2=0.262,P〉0.05;等位基因频率：χ^2=0.141,P〉0.05）。（2）哮喘组儿童血清IL-13水平为（7.89±4.83）ng/L,对照组为（6.06±3.83）ng/L,两组比较差异有统计学意义（P〈0.05）。（3）哮喘组儿童GG、GA、AA基因型血清IL-13水平分别为（7.10±3.46）、（6.40±3.31）、（9.55±5.28）ng/L,组间比较差异均无统计学意义（P〉0.05）。结论 IL-13基因Arg130Gln多态性与新疆维吾尔族哮喘儿童及其血清IL-13水平无关联。血清IL-13水平升高与新疆地区维吾尔族儿童哮喘有关联。Objective To discuss Xinjiang Uygur asthma children IL-13 gene polymorphism and its correlation with the level of serum IL-13.Methods Polymerase chain reaction and direct gene sequencing method was used to analyze the polymorphism of gene IL-13Arg130 Gln in 38 asthma children and 30 normal children,and enzyme linked immunosorbent assay was used to measure serum IL-13,and analyzed the serum IL-13 in 2groups children.Results（1）The gene type and allele gene frequency distribution of gene IL-13 site Arg130Gln in asthma group and control group had no statistical significance（genotype：χ^2=0.262,P0.05;allele frequency：χ^2=0.141,P〉0.05）.（2）The serum IL-13 in asthma group was（7.89±4.83）ng/L and in control group was（6.06±3.83）ng/L,and the difference between two groups was statistically significant（t=1.716,P〈0.05）.（3）The serum IL-13 levels of GG,GA and AA in asthmatic children were（7.10±3.46）,（6.40±3.31）,（9.55±5.28）ng/L,respectively.And there were no significant difference among the groups（P〉0.05）.Conclusion Polymorphism of gene IL-13Arg130 Gln is not associated with asthma of Xinjiang Uygur children and the level of IL-13 in serum.The increasing of serum IL-13 is related to children asthma in Xinjiang area.
