3种南瓜的多糖提取及旋光检测

摘  要:为了探索南瓜中的多糖提取条件和含量测定方法,以3种常见的南瓜为试材,开展了对其多糖提取及旋光检测的试验。采取超声辅助醇沉法提取干、湿南瓜的多糖,并用旋光法测定南瓜多糖的含量。结果表明,干法超声醇沉法南瓜多糖的平均提取率(2.14%)高于湿法(1.07%),干法提取更适于实际应用;采用旋光法测定不同南瓜中多糖含量,金瓜为最高,可达0.33%;长南瓜居中,为0.16%;圆南瓜最低,为0.12%。此法操作简单,快速准确,可应用于南瓜多糖产品的质量检测。In order to study the conditions of polysaccharide extraction from pumpkin and the content determination methods of pumpkin, we carried out the experimental research on the polysaccharide extraction and the polarimetric determination in three types of common pumpkin. The polysaccharide was extracted from the dry and the wet pumpkin respectively by ultra- sonic assisted alcohol sedimentation method. The content was determined by polarimetric method. The results showed that the polysaccharide average extraction percentage of the dry pumpkin (2.14%) was higher than the wet one ( 1.07% ). The dry extraction was more suitable for practical application. The content of polysaccharide in the golden pumpkin(0.33% ) was higher than the others, and long squash was the middle (0.16%), and the round squash was the lowest (0.12%) of them. The polarimetric determination was simple, rapid and accurate for the polysaccharides content, and could be applied to the quality inspection on the pumpkin polysaccharide product.
