摘  要:目的:研究甲嘎松汤对急性肝损伤小鼠的肠道菌群和细胞因子的影响。方法:甲嘎松汤预防给药7 d后,采用四氯化碳制备小鼠急性肝损伤模型。测定肝功能指标;菌落计数法检测盲肠内容物中大肠杆菌、粪肠球菌、双歧杆菌、乳酸杆菌数量,计算B/E值;ELISA法测定血清和肝组织中细胞因子的水平。结果:甲嘎松汤高剂量组ALT水平明显低于模型对照组,甲嘎松汤中剂量组AST水平明显低于模型对照组;甲嘎松汤高剂量组肠道中大肠杆菌数量显著低于模型对照组,甲嘎松汤低、中剂量组肠道中双歧杆菌数量显著高于模型对照组;甲嘎松汤低、中、高剂量组B/E值均显著高于模型对照组;甲嘎松汤低、中、高剂量组IL-6、IFN-γ、TGF-β1水平均明显低于模型对照组,IL-10水平明显高于模型对照组。结论:甲嘎松汤可改善急性肝损伤小鼠肠道菌群失调状态,调节细胞因子的水平,对急性肝损伤小鼠具有保肝作用。Objective: To investigate the impacts of Jiagasong Soup (甲嘎松汤 JGSS) on gut flora and levels of different cytokines in acute liver injury mice. Methods: After treated with JGSS for 7 days, the CCl4-induced acute liver damage model was established. The liver function indexes, number of Escherichia coli, Enterococcus faecalis, Bifidobacterium and Lactobacillus in mice cecum by colony counting, the B/E values and levels of cytokines in serum and liver tissue content by ELISA were detected. Results: Compared with the model group, both ALT levels of JGSS 0.66 g/kg group and AST level of JGSS 0.33 g/kg group were significantly decreased. The ]GSS 0.66 g,/kg group had fewer Escherichia coil colouies than model group, axed much colonies of Bifidobacterium had been observed in JGSS 0.33 g/kg and 0.167 g/kg groups. The B/E values arised dramatically in all JGSS-treated groups. The dramatic down-regulation of IL-6, IFN-% TGF'I31 levels and up-regulation of IL-10 level were also noticed in JGSS-treated groups. Conclusion: The Jiagasong Soup has positive protective effects on liver functions through adjusting alteration of intestinal flora and levels of cytokines in acute liver damage mice.
