摘  要:目的探讨灵芝三萜组分(GLE)增强紫杉醇(PTX)诱导人表皮生长因子受体2阳性(HER2^+)乳腺癌细胞凋亡的作用和机制。方法磺酰罗丹明B(SRB)法和台盼蓝染色计数法检测细胞增殖活性。CompuSyn软件分析药物相互作用。免疫印迹法检测蛋白表达。采用人乳腺癌细胞裸鼠移植瘤模型研究体内抗肿瘤作用。结果体外实验表明,GLE与PTX协同抑制SKBR-3细胞的增殖和诱导细胞凋亡,并协同诱导细胞凋亡相关蛋白的表达和阻断HER2信号通路的传导。体内动物实验表明,GLE增强PTX抗HER2^+乳腺癌的作用。结论GLE与PTX在体内外对HER2^+乳腺癌SKBR-3细胞具有协同抑制作用。Objective To study the enhancement effect of paclitaxel(PTX)induced apoptosis in HER2+ breast cancer cells by the triterpene component of Ganoderma lucidum(GLE)and its mechanism. Methods Cell proliferation was analyzed by SRB and trypan blue exclusion staining method. The expression of proteins was detected by Western blot. Antitumor activity in vivo was studied in SKBR-3 xenograft model. Results In vitro,GLE and PTX had synergistic effect on the proliferation and induced apoptosis in SKBR-3 cells,and on the upregulation of proteins related to cell apoptosis and blocking transduction of HER2 signaling pathway. In vivo,GLE enhanced antitumor activity of PTX against HER2+ breast cancer. Conclusion GLE and PTX have synergistic antitumor effect against the HER2+ breast cancer cells SKBR-3 both in vitro and in vivo.
