摘 要:目的探讨白藜芦醇调控大鼠心力衰竭模型氧化应激相关基因的作用。方法采用腹主动脉缩窄法制备SD大鼠心力衰竭模型,测定心脏重量指数变化,Real-Time PCR定量分析超氧化物歧化酶(SOD)、血红素氧合酶(HO-1)、谷胱甘肽过氧化物酶(GPx)表达;免疫组织化学法检测SOD。结果白藜芦醇干预组和模型组比较,心脏重量指数改善,高剂量组更为显著;心衰大鼠SOD、GPx、HO-1表达水平明显降低(P<0.05或P<0.01),白藜芦醇干预后,SOD、GPx、HO-1表达水平均有所升高,其中以SOD改善明显(P<0.0 5),高剂量组SOD表达甚至高过假手术组。结论白藜芦醇能提高大鼠心力衰竭模型心肌各氧化相关基因的表达,改善心功能。Objective To observe the effect of resveratrol( RES) on the myocardial oxidation related genes in rat models with heart failure( HF). Methods Sprague-Dawley( SD) rat models with HF were established by partially banding abdominal aortic artery. The cardiac fibrosis was evaluated by heart weight index( HWI). The expression of superoxide dismutase( SOD 1),heme oxygenase( HO-1),glutathione peroxidase( GPX) were measured by real-time polymerase chain reaction( RT-PCR). SOD was detected by immunohistochemistry. Results Compared with model group,HWI was significantly increased in RES group,especially RES high dose group. The expression levels of SOD1,GPx,HO-1 were decreased in model group(P〈0. 01 or P〈0. 05). The expression levels of SOD1,GPx,HO-1 were increased in RES group(P〈0. 05),especially SOD. The activity of SOD in RES high dose group was higher than that in sham operation group. Conclusion RES can improve the expression of the myocardial oxidation related genes in rats with heart failure.
