汉黄芩素对人肺癌A549细胞株增殖活性的影响

摘　　要：目的:本实验通过研究汉黄芩素对人肺癌A549细胞株增殖活性的影响,为其机制的研究及临床应用提供理论依据。方法:实验通过MTT法、生长曲线法、检测不同浓度汉黄芩素、不同作用时间对肺癌细胞增殖活性的影响,通过HE染色实验观察汉黄芩素对A549细胞形态学变化。结果:实验显示汉黄芩素对人肺癌细胞株有较强的抑制作用,当药物浓度达到10μg/m L时,增殖抑制作用明显。随着给药时间延长,细胞存活浓度递减。汉黄芩素能够改变肺癌细胞形态,用药后与对照组比较细胞变圆,体积缩小,核质深染,胞质凝集。结论:汉黄芩素能够抑制人肺癌细胞A549增殖,改变细胞的形态,将对肺癌治疗具有一定的指导意义。Objective： To study the effect of wogonin on proliferation of human lung cancer cells A549 to provide theoretical foun- dation for clinical application. Methods：MTT assay and the cell growth curve were adopted to detect the effects of different concentra- tions of Wogonin on the proliferation activity of lung cancer cells. The morphological changes of A549 cells were observed by HE staining. Results： Wogonin could apparently inhibit human lung cancer cell line. When the drug concentration reached 10μg/mL, the proliferation inhibition was obvious. The cell survival concentration decreased with the time.Wogoin could change the shape of A549 cells, such as cell roundeing, volume reduction, nuclear anachromasis, cytoplasm agglutination.Conclusions：Wogonin could inhibit proliferation of human lung cancer cells A549 and will have a certain guiding significance to the treatment of lung cancer.
