摘 要:探讨基于斑马鱼模型的不同浓度的氨基葡萄糖盐酸盐及淫羊藿提取物的促进骨骼矿化活性。选用发育正常的72 hpf斑马鱼胚胎,以培养水为溶剂对照,各受试组分别加入不同浓度(1、10和100μg/m L)的氨基葡萄糖盐酸盐溶液或淫羊藿提取物溶液,连续培养4 d后,用体积分数为0.2%钙黄绿素染色1 h,在体式荧光显微镜下观察斑马鱼骨骼发育情况,拍照并用图像分析软件计算骨骼染色面积。与对照组相比,各受试组对斑马鱼骨骼矿化均呈现一定程度的促进作用,且呈现明显的剂量依赖性。氨基葡萄糖盐酸及淫羊藿提取物均具有较好的促进斑马鱼骨骼矿化的活性。相同浓度条件下,氨基葡萄糖盐酸盐较淫羊藿提取物具有更好的促进骨骼矿化活性。We addressed the activity of bone mineralization promotion of different concentrations of glucosamine hydrochloride and epimedium extract based on zebrafish model. We selected 72 hpf normal growth zebrafish embryos as subjects, and cultivation water as control group or different concentrations (1, 10 and 100 μg/mL) of glucosamine hydrochloride solutions or epimedium extract solutions as experiment group. After four-day cultivation, all the zebrafish was dyed for 1 h with 0.2% calcein. We then observed their skeleton growth with a microscope, and calculated its stained area through an image analysis software. Results indicate that glucosamine hydrochloride and epimedium extract of all experiment groups all have some promotion effect on skeleton mineralization of zebrafish, as compared with control group, obvious dose dependence. This shows that both glucosamine hydrochloride and epimedium extract have significant bone mineralization activity to zebrafish. Moreover, glucosamine hydrochioride has better mineralization activity than epimedium extract for the same concentration.
