摘  要:目的探讨叶酸对人脂肪间充质干细胞(hADSCs)成脂分化过程的影响及机制。方法以正常叶酸(Control)和无叶酸(FD)成脂诱导液对hADSCs细胞进行成脂诱导,不同时间分别采用油红O染色观察细胞内脂滴生成情况;采用SYBR Green实时荧光定量RT-PCR方法检测成脂分化相关转录调控基因PPARγ和C/EBPα的表达;采用蛋白质印迹法检测PPARγ蛋白的表达。结果与正常叶酸组相比,无叶酸组脂滴数量明显减少;两组细胞成脂分化相关转录调控基因PPARγ和C/EBPαmRNA表达无明显差异;PPARγ蛋白表达明显低于正常叶酸组。结论叶酸可通过上调PPARγ的表达而促进hADSCs细胞成脂分化。Objective To study the effect and mechanism of folate on the adipogenesis process of human adipose derived mesenchymal stem cells(h ADSCs). Methods h ADSCs were cultured in normal and folate-deficient adipogenic induction medium(control group or folate-deficient group). Lipid droplets were measured by oil red O staining. Nuclear transcription factors PPARγ and C/EBPα expression were measured by real time q PCR. The expression of PPARγ was measured by western blot. Results Compared with the normal group, there was an evident decrease of lipid droplets in the folate- deficient group. The expression of PPARγ and C/EBPα showed no significant difference in m RNA level, but the expression of PPARγ protein is down-regulated in folate-deficient group. Conclusion Folate can promote the adipogenesis process of h ADSCs by upregulating the expression of PPARγ.
