摘  要:目的观察过表达miR-126的骨髓间充质干细胞心肌内注射对急性心肌梗死后大鼠心脏功能的改善情况。方法制备过表达miR-126的骨髓间充质干细胞(MSCmiR-126),通过冠脉结扎的方法制作大鼠急性心肌梗死模型,制备成功的模型随机分为三组:MSCmiR-126组(n=25)、骨髓间充质干细胞(MSC)组(n=25)及磷酸缓冲盐(PBS)对照组(n=25),各组动物分别于心肌梗死区注射相应细胞或试剂,6周后行超声心动图检查观察各组动物心脏功能情况,而后处死动物取出心脏,通过组织学方法观察各组动物心室重构的差异。结果 MSCmiR-126组超声心动图及组织学指标均优于MSC组,这两组又都优于PBS组,差异均具有统计学意义。结论移植经miR-126修饰的骨髓间充质干细胞能够明显改善大鼠心肌梗死后的心功能,效果优于普通骨髓间充质干细胞移植。Objective Observe the condition of cardiac function post intramyocardium injection with the bone mesenchymal stem cells( MSC) overexpressed miR-126. Methods Prepared the bone MSC Cells overexpressed miR-126( MSCmiR-126),establish the rat models of acute myocardial infarction by the method of coronary artery occlusion. Then divided these models into three groups: MSCmiR-126 group( n = 25) 、MSC group( n = 25) and PBS control group( n = 25). Each group was injected with corresponding cells or reagents at area of myocardial infarction. Meatured the condition of cardiac function by ultrasound cardiogram 6 weeks later,then sacrificed models to collect heart,investigated the difference of ventricular remodeling among groups by histology method. Results In MSCmiR-126 group the index of ultrasound cardiogram and histology superior to MSC group,and both groups superior to PBS control group,there was significantly differences with statistically. Conclusion Transplant the bone MSC modified with miR-126 could improve the cardiac function after myocardial infarction,and the effect superior to ordinary bone MSC transplantation.
