摘  要:目的初步探讨二甲基亚砜(DMSO)对正常与高糖刺激下大鼠肾小球系膜细胞(RMCs)生长、形态的影响。方法根据细胞培养基中葡萄糖含量及DMSO浓度的不同,采用成组设计方法随机将细胞分为NG组(给予葡萄糖浓度为5.6 mmol/L低糖DMEM完全培养基)、MG组(给予甘露醇浓度为25 mmol/L DMEM完全培养基)、高糖组(HG,给予葡萄糖浓度为25 mmol/L DMEM完全培养基)、DMSO+NG组(给予含0.1%、0.5%、1.0%、1.5%、2.0%、2.5%、3.0%、3.5%DMSO的低糖DMEM培养基)、DMSO+HG组(给予含0.1%、0.5%、1.0%、1.5%、2.0%、2.5%、3.0%、3.5%DMSO的高糖DMEM培养基),以细胞存活率为考察指标,采用MTT法检测不同处理组细胞活力。结果与NG组比较,当DMSO终浓度大于0.5%时,DMSO+NG组细胞形态改变,细胞活力有显著性变化,差异有统计学意义(P〈0.01)。与HG组比较,当DMSO终浓度大于0.1%时,DMSO+HG组细胞形态改变,细胞活力发生极显著性变化,差异有统计学意义(P〈0.01)。在0.5%-2.5%浓度范围内,DMSO+NG组与含相同体积的DMSO+HG组比较,差异有统计学意义(P〈0.01)。DMSO对正常生长环境下细胞的半数抑制率为2.402%,对高糖刺激条件下细胞的半数抑制率为2.114%。结论基于DMSO对高糖刺激下大鼠RMCs细胞影响更为敏感,可为DMSO作溶剂进行同类体外活性筛选提供一定参考。Objective To investigate the effects of dimethyl sulfoxide(DMSO) on growth and morphology in rats' renal mesangial cells(RMCs) in high glucose and normal medium. Methods According to the concentration of glucose and DMSO in the cell culture medium and using the method of group design, the cells were randomly divided into NG group(the concentration of glucose was 5.6 mmol/L), MG group(the concentration of mannitol was 25 mmol/L), high glucose group(HG, the concentration of glucose was 25 mmol/L), DMSO and NG group(the concentration of DMSO was0.1%, 0.5%, 1%, 1.5%, 2.0%, 2.5%, 3.0% and 3.5% in the normal group) and DMSO and HG group(the concentration of DMSO was 0.1%, 0.5%, 1.0%, 1.5%, 2.0%, 2.5%, 3.0% and 3.5% in the high glucose group). Cell viability was selected as evaluation index and carried out with colorimetric assay with MTT. Results Compared with NG group, when the DMSO concentration was higher than 0.5%, the morphous of DMSO and NG group cells changed and the cell viability had statistical difference(P〈0.01). Compared with HG group, when the DMSO concentration was higher than0.1%, the morphous of DMSO and HG group cells changed and the cell viability had statistical difference(P〈0.01).DMSO and HG group had significant difference compared with DMSO and NG group, which had same concentration of DMSO, when the concentration of DMSO was changed between 0.5% and 2.5%(P〈0.01). The half of the inhibition of DMSO for normal cells was 2.402%, and for cells induced by high glucose medium was 2.114%. Conclusion Based on the fact that the cells induced by high glucose medium were more sensitive, it can provide reference for DMSO as a solvent mediator to the similar activity screening in vitro.
