摘 要:目的:观察六味地黄汤对Ig A肾病大鼠外周血C1GALT1及Cosmc mRNA表达,探讨其对Ig A糖基化的影响。方法:SD大鼠40只随机分为正常对照组,模型组,雷公藤多苷片组,六味地黄汤组。除正常对照组外均采用运用牛血清白蛋白+脂多糖+四氯化碳方法建立实验性Ig A肾病模型。造模成功后连续灌胃4周。将各组大鼠外周血分离的PBMC悬液,采用实时荧光定量PCR检测C1GALT1及Cosmc mRNA的表达,ELISA法测定外周血中Ig A含量,凝集素亲和ELISA法检测Ig A1异常糖基化程度,并进行定量分析。结果:各组C1GALT1mRNA表达量并差异无统计学意义(P>0.05);在各组Cosmc mRNA表达中,六味地黄汤组及雷公藤多苷片组表达均较模型组升高(P<0.05),但六味地黄汤组表达较雷公藤多苷片组低(P<0.05);在各组大鼠Ig A含量及异常糖基化程度比较中,六味地黄汤组较模型组下降,差异具有统计学意义(P<0.05)。结论:六味地黄汤可以上调Cosmc mRNA表达,增加C1GALT1活性,改善Ig A1糖基化异常,从而下调体内Ig A1含量,避免进一步沉积肾脏,延缓Ig A肾病的进展。Objective: To observe the effects of Liuwei Dihuang Decoction on Ig A nephropathy rat peripheral blood C1GALT1 and Cosmc mRNA expression,and to investigate its effect on Ig A glycosylation. Methods: Forty SD rats were equally randomized into 4 groups: the normal group,model group,tripterygium glycosides group,Liuwei Dihuang decoction group. Ig AN rat model was established with bovine serum albumin( BSA),lipopolysaccharide( LPS),carbon tetrachloride( CCl4). After 4 weeks of continuous perfusion. Each group of rat peripheral blood separation of PBMC suspension,using real- time fluorescent quantitative PCR detection C1GALT1 and Cosmc mRNA expression,ELISA method for determination of Ig A in peripheral blood content of lectin affinity ELISA method to detect abnormal Ig A1 glycosylation,and carry on quantitative analysis. Results: Each C1GALT1 mRNA expression quantity is no significant difference( P〉0. 05); In each group Cosmc mRNA expression,Liuwei Dihuang decoction group and tripterygium glycosides tablets more expression were higher in the model group( P〈0. 05),but express Liuwei Dihuang decoction group lower than tripterygium glycosides tablets group( P〈0. 05). The Ig A content and the degree of abnormal glycosylation of the rats in each group were compared. The Liuwei Dihuang decoction group were significantly lower than the model group,the difference was statistically significant( P〈0. 05). Conclusion: Liuwei Dihuang decoction can regulate the expression of Cosmc and mRNA,increase the activity of C1GALT1,improve the Ig A1 abnorant glycosylation,which down regulate the Ig A1 content,to avoid further deposition in kidney,and delay the progression of Ig A nephropathy.
