摘  要:目的:明确VEGF抗体在人肾小球系膜细胞高糖状态模型生长、增殖和迁徙中的重要作用,为VEGF抗体应用于糖尿病肾病的临床治疗提供实验依据。方法:(1)建立人肾小球系膜细胞的高糖状态模型;(2)检测人肾小球系膜细胞高糖状态模型自分泌的VEGF;(3)观察VEGF抗体对人肾小球系膜细胞高糖状态模型生长、增殖和迁徙能力的影响。结果:(1)与对照组、低糖组比较,高糖组的肾小球系膜细胞分泌VEGF量最多,差异有统计学意义(P<0.05);(2)在人肾小球系膜细胞高糖状态模型的培养基中加入不同浓度的VEGF抗体,随VEGF抗体浓度升高,其细胞凋亡率、细胞增殖并非呈线性关系;(3)在人肾小球系膜细胞高糖状态模型中提取细胞,加入transwell上层小室,加入不同浓度的VEGF抗体,VEGF抗体浓度一定范围内增高会降低系膜细胞迁徙能力。结论:(1)高糖引起的肾小球系膜细胞VEGF表达上调,可能是糖尿病肾病的发病机制之一;(2)VEGF抗体促进肾小球系膜细胞的凋亡,降低细胞增殖能力与迁徙能力。Objective: We observed the expression of VEGF secretion,cell growth,proliferation and migration in human glomerular mesangial stimulated by high glucose; and the function of VEGF antibodies. Methods:( 1) Establishment of human mesangial cells in high glucose condition model;( 2) Detected in human mesangial cells high glucose condition model autocrine VEGF;( 3) Observed effects of VEGF antibody human mesangial cell growth,proliferation and migration ability of models. Results:( 1)VEGF respectively detected by ELISA obtained under high glucose stimulation,VEGF secretion highest,there was a significant statistical difference( P〈0. 05) with the normal group;( 2) With VEGF antibody concentration,at the same time stimulating their apoptosis rate is not a linear relationship,but the apoptosis rate increased initial concentration,but to a certain concentration( 100 pg / ml)occurs when a plateau. Similarly,cell proliferation,apoptosis is also similar with the initial concentration with VEGF,decreased cell proliferation,but to a certain concentration( 100 pg / ml),cell proliferation is not the case there continues to decline;( 3) The human mesangial cellshigh glucose conditionmodel added 8μm pore transwell upper chamber,while adding different concentrations of VEGF antibody,12 h after counting the number of cells into the lower chambers. The experiment used a three density levels found in the antibody concentration range increased VEGF reduces mesangial cell migration capacity. Conclusion:( 1) VEGF expression in mesangial cells leads to upregulation of high- sugar,may be one of the pathogenesis of diabetic nephropathy;( 2) VEGF antibodies induce apoptosis in glomerular mesangial cells,reducing cell proliferation and migration capabilities.
