摘 要:目的探讨白花丹参水提物对糖尿病(DM)大鼠心肌细胞凋亡的影响。方法选择48只大鼠予高糖高脂饲养,经尾静脉注射链脲佐菌素35 mg/kg制备DM模型,随机分为DM组、LD组、HD组、WD组,每组各12只。另选择12只正常大鼠作为正常对照组(NC组)。造模成功后,LD组、HD组分别以白花丹参提取物15、30 g/(kg·d)灌胃,WD组以厄贝沙坦17.5 mg/(kg·d)灌胃,DM组以等剂量的生理盐水灌胃,均持续4周。NC组不灌胃。各组于造模后第4周末,末次给药后处死,取心肌组织,采用Western blot法检测凋亡相关蛋白B淋巴细胞瘤-2(Bcl-2)蛋白、Bcl-2相关X(Bax)蛋白、Bcl-2/腺病毒E1B19k D相互作用蛋白3(Bnip-3)的表达。结果 DM组细胞凋亡明显,WD组、LD组细胞凋亡较DM组减轻,HD组心肌病理表现明显好于WD组和HD组。DM组Bcl-2蛋白表达低于其他各组,WD组、LD组Bcl-2蛋白表达均低于HD组(P均<0.01)。DM组Bax、Bnip-3蛋白表达均高于其他各组,WD组、LD组Bax、Bnip-3蛋白表达均高于HD组(P均<0.01)。结论白花丹参可减轻DM大鼠心肌细胞的凋亡,其机制可能与增加心肌组织Bcl-2蛋白表达、降低Bax及Bnip-3蛋白表达有关。Objective To investigate the effects of Salvia miltiorrhiza water-extract on apoptosis of myocardial ceils in rats with diabetes mellitus (DM). Methods Forty-eight male Wistar rats were fed with high-glucose and high-fat diet and then were injected with streptozotocin (35 mg/kg) to establish diabetes mellitus (DM) models. After that, all model rats were randomly divided into the DM group (DM group) ,'Western medicine group (WD group), low-dose (LD) and highdose (HD) Chinese medicine groups, 12 rats in each group. Meanwhile, another 12 healthy rats were selected as the normal control group (NC group). After modeling, LD and HD groups were given 15 g/( kg · d) and 30 g/( kg · d) Salvia miltiorrhiza water-extract through intragastric administration, WD group was given 17.5 mg/(kg · d) irbesartan, and DM group was treated with the same dose of normal saline for 4 weeks. On week 4 after modeling, the expression of apoptosis-related protein ( Bcl-2), Bcl-2-related protein (Bax) and Bnip-3 was detected by Western blotting. Results The apoptosis of myocardial cells in the DM group was more significant, the apoptosis of the WD and LD groups alleviated as compared with that of the DM group, and the pathological manifestations of the HD group was better than that of the WD and LD group. The Bcl-2 protein expression of the DM group was lower that of the other groups, and the Bcl-2 protein expression of the WD and LD groups was lower than that of the HD group (all P 〈0.01 ). Bnip-3 and Bax protein expression of the DM group was higher than that of the other groups, and Bnip-3 and Bax protein expression of the WD and LD groups was higher than that of the HD group ( all P 〈 0. 01 ). Conclusion Salvia miltiorrhiza water-extract can alleviate the apoptosis of myocardial cells in DM rats, and the mechanism may be related to the increased Bcl-2 protein expression and the decreased Bnip-3 and Bax protein expression.
