摘 要:目的通过观察变应性鼻炎大鼠鼻黏膜、支气管和肺组织形态学变化和细胞因子的阳性表达,探讨鼻渊舒口服液治疗变应性鼻炎(Allergic rhnitis,AR)的机制。方法样本分为四组,I组(空白对照组n=10)、II组(模型组n=10)、III组(阳性对照组n=10)、IV组(鼻渊舒组n=10),I组用生理盐水致敏并灌胃,II、III、IV组用卵清蛋白致敏大鼠,制成变应性鼻炎动物模型,II组用生理盐水灌胃,III组用地塞米松腹腔注射,IV组将鼻渊舒口服液灌胃给药,应用免疫组化和原位杂交观察IL-5、IL-13、在鼻黏膜、支气管、肺组织中的表达,同时观察鼻黏膜、支气管、肺组织中的表达,同时观察鼻黏膜、支气管和肺组织形态学变化。结果鼻渊舒组IL-5、IL-13阳性表达均低于模型组,其差异有显著性意义(P<0.01),模型组鼻黏膜、支气管、肺组织中有大量嗜酸性细胞浸润,鼻渊舒组及空白对照组较少有上述改变。结论鼻渊舒口服液可以抑制变应性大鼠鼻黏膜、支气管、肺组织中IL-5、IL-13的阳性表达,并抑制炎症细胞在气道粘膜中的聚集。Objective To research the mechanism of Nasosinusitis Relieving Dral Liquid remedying allergic rhinitis by the meanof observing morpholigical transformation and positive expression of cytokine in the nose mucosa/bronchus and lung tissue in Allergicrhinitis rat. Mrthods the sample is divided into four groups,separately,I group(the blank contrast group n=10)II group(the modelgroup n=10)III group(positive contrast group n=10)IV group(the therapy group n=10)I group are done with normal saline sensitiza-tion and pouring into the stomach II、III、IV groups are done with the ovalbumin sensitizating the rat and execute the Allergic rhinitisanimal model.Theanimalsof II group are poured into the stomach with normal saline,The animals of III group are injected into the abdo-men with the dexamethasone,The animals of IV group are poured into the stomch with Nasosinusitis Relieving Dral Liquid and thenobserving the positive expression of the IL-5、IL-3 in the nose mucosa、bronchus and lung tissue by the way of the immunization andthe hybridization in situ,simultaneously,observing morphological transformation in the nose mucosa、bronchus and lung tissue.Results Serum IL-5、IL-13 in the rapy group are lower than that inmodel grouptha(tP0.05);Under light microscope of nose mucosa inallergic rhinitis model,there are a lot of eosinophils and mase cells infiltration and there is mast cells degranulation,In bland contrastgroup and therapy group,there is less eosinophils.Conclution this result suggests that Nasosinusitis Relieving Dral Liquid could de-press the positive expression of cytokine IL-5、IL- in the nose mucosa、bronchus and lung tissue in allergic rhinitis rat and depress theconcentration of inflammation cells in air duct mucosa.
