摘 要:目的探讨不同浓度布洛芬对体外细粒棘球蚴原头节生长的抑制作用。方法实验分为空白对照组、DMSO组和布洛芬处理组(0.5、1.0、2.0、4.0mmol/L),将体外培养的细粒棘球蚴原头节加入相应培液中孵育,经0.1%伊红染色后在倒置显微镜下观察原头节形态及活力,实验重复3次;扫描电子显微镜(SEM)下观察原头节的超微结构变化;孵育24h后用caspase-3活性检测试剂盒检测原头节caspase-3酶活性。结果空白对照组和DMSO组原头节活力无明显变化。2.0mmol/L和4.0mmol/L布洛芬组作用48h后原头节活力开始下降,作用12d后4.0mmol/L布洛芬处理组无存活的原头节,2.0mmol/L布洛芬处理组的原头节活力为26.89%,0.5mmol/L和1.0mmol/L组原头节活力也明显下降。SEM观察超微结构,4.0mmol/L布洛芬组处理3d后的原头节顶突外翻、变形,吸盘变形,虫体出现虫蛀样损害。布洛芬处理组作用24h后caspase-3酶活性分别为(18.486±0.450)、(29.045±0.273)、(36.203±0.450)、(47.537±0.450)×103活力单位,对照组为(10.016±0.358)×103活力单位,差异有统计学意义(F=3177.897,P<0.05)。结论布洛芬对体外细粒棘球蚴原头节有抑制作用。Objective To investigate the inhibition of Echinococcus granulosus protoscoleces by ibuprofen in vitro.Methods Protoscoleces were cultured in vitro in the presence of RPMI 1640 medium or treated with DMSO or ibuprofen at concentrations of 0.5,1.0,2.0,and 4.0 mmol/L.Changes in morphology and viability were investigated with the help of 0.1% eosin staining and light microscopy.The experiment was repeated three times.Ultrastructural changes in E.granulosus protoscoleces due to ibuprofen were visualized with scanning electron microscopy(SEM).Caspase-3was detected in protoscoleces using a caspase-3activity assay kit. Results After 12 days,control protoscoleces and protoscoleces treated with DMSO remained viable and had no marked changes.In contrast,treatment with 2.0or 4.0mmol/L of ibuprofen reduced the number of viable protoscoleces within a period of 2days.After 12 days,protoscoleces treated with 4.0mmol/L of ibuprofen were no longer viable.At 2.0mmol/L,ibuprofen had a markedly reduced efficacy,with26.89% of protoscoleces still viable.Changes in viability were marked when protoscoleces were treated with lower concentrations of ibuprofen.Ultrastructural changes were investigated using SEM.No ultrastructural damage was observed in protoscoleces that were not treated with ibuprofen during the period of the study.In contrast,ultrastructural changes were detected in protoscoleces treated with ibuprofen.Changes included contraction of the soma region,formation of blebs,rostellar disorganization,loss of hooks,and destruction of microtriches.Caspase-3was detected in control protoscoleces and treated protoscoleces,and the activity of this enzyme was higher 24 hafter protoscoleces were treated with ibuprofen. Conclusion This study indicates that ibuprofen is a potential protoscolicidal agent against E.granulosus.
