益母草试管苗愈伤组织培养及其生物碱含量的测定

摘  要:以种子萌发的试管苗叶片为外植体,采用正交试验探究了其愈伤组织的形成,并测定了其生物碱含量。结果表明:1.5 mg/L 6-BA利于种子萌发,其萌发率最高达26.39%;叶片愈伤组织诱导适宜培养基组合为MS+2.0 mg/L 6-BA+0.5 mg/L 2,4-D+20 g/L蔗糖+7 g/L琼脂,诱导率可达80.00%;适宜愈伤组织增殖激素组合为2.0 mg/L 6-BA+0.3 mg/L 2,4-D,增殖率可达88.05%,但各激素组合对愈伤组织生物碱的含量并无显著性差异。The effects of different 6- BA concentrations on the motherwort seeds′ germination werestudied,and the leaves of plantlets which generated from the seeds,were selected as the explants,toexplore the suitable conditions of callus induction,using the orthogonal design,the callus′ proliferationconditions and alkaloid content were studied. The results showed that when the concentration of 6-BA was1. 5 mg/L,the seeds′ germination rate was highest,up to 26. 39%;the suitable medium composition forcallus′ induction was MS + 6-BA 2. 0 mg/L + 2,4-D 0. 5 mg/L + sucrose 20 g/L + agar 7 g/L,inductionrate reached 80. 00%;the suitable hormone combinations for the callus′ proliferation was 2. 0 mg/L 6-BA+0. 3 mg/L 2,4-D,the proliferation rate reached 88. 05%,but the above hormone combinations oncallus′ alkaloid content had no significant difference.
