摘  要:目的:优化贡菊不定芽诱导与增值的培养条件,为建立和完善贡菊离体快繁体系奠定基础。方法:以贡菊无菌组培苗带芽茎段为试材,通过单因素、组合、正交试验等方法探讨不同植物生长调节剂对不定芽诱导、增值的影响,并对基本培养基进行优化。结果:优选出适用于不定芽诱导的细胞分裂素为6-BA,本实验条件下不定芽诱导最佳配方为:MS+6-BA2.0 mg·L-1+NAA0.1 mg·L-1或6-BA2.0-1mg·L-1+IBA0.1 mg·L-1;不定芽增值最佳配方为:MS+6-BA2.0 mg·L-1+NAA0.02 mg·L-1+IBA0.1 mg·L-1;基本培养基优化结果为:大量元素MS标准浓度+铁盐1.5倍MS标准浓度+蔗糖20 g·L-1+椰子汁30 m L·L-1。结论:6-BA对不定芽诱导有显著影响,IBA、NAA对不定芽诱导增殖具有一定调节作用;基本培养基的改良优化可增强不定芽生长发育。Objective: The culture conditions of adventitious buds induction and proliferation were optimized to lay a foundation for establishing and perfecting the rapid propagation system of Dendranthema morifolium. Methods: Single factor test,combination test and orthogonal test were used,also the compositions of basic medium were optimized to investigate the effect of different plant growth regulator on adventitious buds induction and proliferation. Results: The results showed that the optimum cytokinin for induction was 6-BA; the best medium for induction was MS + 6-BA2. 0 mg·L- 1+ NAA0. 1 mg·L- 1or 6-BA2. 0 mg·L- 1+ IBA0. 1 mg·L- 1; the best medium for propagation was MS + 6-BA2. 0 mg·L- 1+ NAA0. 02 mg·L- 1+IBA0. 1 mg·L- 1; the optimized basic medium compositions were: basic MS macronutrients concentration + 1. 5 times Fe +sucrose 20 g·L- 1+ coconut water 30 ml·L- 1. Conclusion: 6-BA has obvious effect,and also IBA and NAA had regulating effect on induction and propagation of adventitious beds,in addition,the improved medium could promote adventitious buds growth.
