摘  要:目的:观察艾灸对溃疡性结肠炎大鼠结肠Toll样受体4(TLR4)和肿瘤坏死因子α(TNF-α)蛋白及其m RNA表达的影响,进一步探讨艾灸治疗溃疡性结肠炎的作用机制。方法:将40只SD大鼠随机分为正常组、模型组、隔药灸组和柳氮磺胺吡啶(Sulfasalazine,SASP)组。采用隔药灸天枢和气海穴及SASP灌胃进行干预。干预结束后,应用HE染色光镜下观察各组大鼠结肠病理学变化,分别采用Western blot法和FQ-PCR法检测大鼠结肠TLR4和TNF-α蛋白及其m RNA表达。结果:造模大鼠存在结肠组织病理学损伤,隔药灸组和SASP组大鼠结肠病理学与模型组相比均有一定的改善;与正常组比较,模型组大鼠结肠TLR4和TNF-α蛋白及其m RNA表达均显著增加(P<0.05);经隔药灸和SASP干预后,大鼠结肠TLR4和TNF-α蛋白及其m RNA表达均显著降低(P<0.05)。结论:TLR4、TNF-α可能在溃疡性结肠炎中发挥炎症损伤作用,隔药灸能下调溃疡性结肠炎大鼠结肠TLR4和TNF-α的表达,提示其机制可能是通过下调TLR4和TNF-α表达介导的级联反应达到艾灸治疗溃疡性结肠炎的目的。This study was aimed to observe the effect of herb-partition moxibustion on the protein and m RNA expressions of Toll-like receptor 4(TLR-4) and tumor necrosis factor-α(TNF-α) in rats with ulcerative colitis(UC), and to further explore the mechanism of herb-partition moxibustion on UC. A total of 40 male SD rats were randomly divided into four groups: the normal group, the model group, the herb-partition moxibustion group and the sulfasalazine(SASP) group. Herb-partition moxibustion was used on Tianshu and Qihai acupoint in the herb-partition moxibustion group, and SASP gavage was used in the SASP group. The colonic pathological change was detected by HE staining. Protein and m RNA expression of TLR-4 and TNF-α were detected by western blot and FQ-PCR methods, respectively. The results showed that pathological damages appeared in colon tissues of the model rats. Compared with the model group, the colon pathological damage was improved in the herb-partitioned moxibustion group and the SASP group. Protein and m RNA expression of TLR-4 and TNF-α were significantly increased in the model group rats compared with the normal group(P〈 0.05). After herb-partitioned moxibustion and SASP treatments, the protein and m RNA expression of TLR-4 and TNF-α were significantly decreased(P〈 0.05). It was concluded that TLR-4 and TNF-α may play an important role in inflammatory injury of UC. Herb-partitioned moxibustion can reduce the expression of TLR-4 and TNF-α. It suggested that the underlying mechanism may be the down-regulation of the cascade reaction mediated by TLR4 and TNF-α expression to achieve the purpose of moxibustion to treat UC.
