摘  要:目的:建立连二亚硫酸钠(Na2S2O4)诱导SH-SY5Y神经细胞缺氧缺糖损伤模型,进行提取物筛选,对筛选出的有活性提取物初步探讨其对神经细胞的保护作用。方法:CCK-8检测细胞存活率,比色法检测乳酸脱氢酶(LDH)释放,超氧化歧化酶活性(SOD),丙二醛含量(MDA),荧光探针负载法检测细胞内活性氧(ROS)和细胞内钙离子([Ca2+]i)荧光强度。结果:连二亚硫酸钠诱导SH-SY5Y细胞损伤模型上进行提取物初筛,发现泽泻白术药对提取物有一定活性。与模型组比较,提取物组细胞的LDH释放降低,SOD活性升高,MDA含量降低,细胞内ROS水平和[Ca2+]i荧光强度均减弱。结论:泽泻白术药对提取物对连二亚硫酸钠致SH-SY5Y神经细胞损伤有一定的脑保护作用。This study was aimed to establish the Na2S2O4 induced SH-SY5 Y cell in order to screen model for the research of effective extractions of herb pairs and its related mechanism. CCK-8 was used to detect cell survival rate. Colorimetric method was used to detect the releasing of lactate dehydrogenase(LDH), superoxide dismutase(SOD) and malondialdehyde(MDA). Fluorescence probe method was used to detect the fluorescence intensity of reactive oxygen species(ROS) and intracellular calcium concentration([Ca^2+]i). The results showed that the extract screened with Na2S2O4 induced SH-SY5 Y cell indicated that herb pair of Zexie-Baizhu had certain activity on the extract. Compared to the model group, the releasing of LDH decreased, SOD activity increased, the content of MDA decreased, and the fluorescence intensity of ROS and [Ca^2+]i decreased in the extract group. It was concluded that herb pair of Zexie-Baizhu had certain cerebral protective effects on Na2S2O4 induced SH-SY5 Y cell.
