摘  要:目的:应用效应面法及药效筛选法优选鼻鼽颗粒的提取工艺。方法:采用效应面法的方法,以迷迭香酸提取率为指标,通过单因素试验和Box-Behnken效应面法考察提取溶剂量、提取时间、提取次数对提取工艺的影响,进一步通过药效筛选确定鼻鼽颗粒的疗效。结果:与模型组比较,鼻鼽颗粒工艺血清细胞因子水平均有下降趋势,可降低Ig E和IFN差异有统计学意义(P<0.05,P<0.01)。最佳提取工艺为提取溶剂量2 430 m L(9倍量),提取2次,每次1.5 h。迷迭香酸提取率84.89%。结论 :鼻鼽颗粒在降低免疫因子水平方面疗效明确,通过综合评价提取物的治疗效果,优选的鼻鼽颗粒工艺提取工艺稳定、可行。This study was aimed to optimize the extraction process of the Biqiu granules by response surface methodology(RSM) and activity screening method. With the rosemary acid extraction rate as index, the solvent volume, extraction duration and extraction times were investigated with the single factor assays and Box-Behnken method. The activity screening method was further utilized to determine the curative effect of Biqiu granules. The results showed that compared with the model group, the Biqiu granules had the descending trend on serum cytokines(Ig E and IFN) with statistical significance(P〈 0.05, P〈 0.01). The optimal extraction process of Biqiu granules was shown as follows: solvent volume 2430 ml(9-fold), extraction duration for 1.5 h, twice. The rosemary acid extraction rate was 84.89%. It was concluded that Biqiu granules had specific curative efficacy on reducing the levels of immune factors and its optimal extraction process was stable and feasible.
