摘 要:目的:研究活血开窍方对转基因AD模型小鼠IL-1、IL-6及TNF-α的影响,探讨其作用机制。方法:3月龄APP swe/PS1d E9双转基因(APP转基因)AD模型小鼠,随机分为活血开窍大剂量组、活血开窍小剂量组、盐酸多奈哌齐组、模型组,C57BL/6J小鼠为正常对照组,每组10只,连续灌胃给药3个月,采用Morris水迷宫实验观测APP转基因AD模型小鼠学习记忆能力的变化,ELISA测定小鼠血清中IL-1、IL-6及TNF-α含量。结果:Morris水迷宫实验结果表明,与正常对照组相比,3月龄模型组小鼠穿越原平台位置的次数、第四象限游泳时间及路程均显著缩短和减少(P<0.05);药物治疗后,小鼠在Morris水迷宫中穿越原平台位置次数、第四象限游泳时间及路程均较模型组显著提高(P<0.05)。ELISA实验结果表明,与正常对照组相比,模型组小鼠血清中IL-1、IL-6及TNF-α含量显著升高(P<0.01);与模型组相比,活血开窍组小鼠IL-1、IL-6及TNF-α血清含量显著降低(P<0.05)。结论:活血开窍方可在一定程度上改善APP转基因AD模型小鼠学习记忆能力,并且通过降低APP转基因AD模型小鼠血清IL-1、IL-6、TNF-α的含量改善其炎性状态。Objective:To study the effects of Huoxue Kaiqiao prescription on cytokines IL-I,IL-6 and TNF-α in the transgenic AD models and explore its action mechanism. Methods:The APP transgenic mice 3 months age were randomly divided into the large dose group of Huoxue Kaiqiao,small dose group of Huoxue Kaiqiao and donepezil hydrochloride group and model group. C57BL/6J mice were selected as the normal control group,with 10 mice in each group,continuous intragastric administration for 3 months. Morris water maze was used to observe the learning and memory abilities of APP transgenic mice. The levels of TNF-α, IL-1 and IL-6 in the serum of mice were detected by ELISA. Results:In the Morris water maze,compared with the normal control group,the times across the platform were reduced and the swimming time and dis- tance in the fourth quadrant were shortened in the model group(P〈0.05). After treatment,compared with the model group,the times across the platform and the swimming time and distance in the fourth quadrant were improved in the Huoxue Kaiqiao group(P〈0.05). ELISA results showed that the levels of IL-1,IL-6 and TNF-α were improved significantly compared with the normal control group(P〈0.01 ). Compared with the model group,the serum levels of IL-1,IL-6,TNF-et in Huoxue Kaiqiao group were significantly reduced after treatment(P〈0.05). Conclusion:Huoxue Kaiqiao prescription can improve the learning and memory abil- ities to some extent and improve the AD model inflammatory state by reducing the levels of IL-1,IL-6, TNF-α in the APP transgenic mice.
