HPLC法同时测定复方米诺地尔凝胶中米诺地尔和维A酸的含量

摘　　要：目的:建立HPLC法同时测定复方米诺地尔凝胶剂中的米诺地尔和维A酸含量。方法:色谱柱:Shim-Pack VP-ODS C18柱(250 mm×4.6 mm,5μm);流动相:甲醇和2%醋酸溶液进行梯度洗脱;检测波长:280 nm和350 nm;流速:1.0 ml·min^(-1);柱温:35℃;进样量:10μl。结果:米诺地尔和维A酸分别在4.0~240.0μg·ml^(-1)和0.05~3.00μg·ml^(-1)的浓度范围内与峰面积呈良好的线性关系(r分别为0.999 6和0.999 3)。米诺地尔和维A酸的平均回收率分别为99.56%和99.36%,RSD分别为0.42%和0.50%(n=9)。结论:建立的测定方法简便,可行,适用于同时测定复方米诺地尔凝胶剂中米诺地尔和维A酸的含量。Objective：To establish an HPLC method for the simultaneous determination of minoxidil and tretinoin in compound minoxidil gel.Methods：A Shim-Pack VP-ODS C_（18）（250 mm × 4.6 mm,5 μm） column was used with the mobile phase of methanol and 2% acetic acid solution by gradient eleution.The detection wavelength was respectively set at 280 nm and 350 nm,and the flow rate was 1.0 ml · min^-1.The column temperature was 35℃ and the injection volume was 10 μl.Results：Minoxidil and tretinoin had a good linear relationship within the range of 4.0-240.0 μg ·ml^-1and 0.05-3.00 μg · ml^-1（r =0.999 6 and 0.999 3）,respectively.The average recovery was 99.56%（RSD = 0.42%,n = 9） and 99.36%（RSD = 0.50%,n = 9）,respectively.Conclusion：The established method is simple,and suitable for the simultaneous determination of the two constituents in compound minoxidil gel.
