摘 要:目的探讨IL-4基因甲基化在支气管哮喘(简称哮喘)患儿和健康儿童中的表达差异及对IL-4、IgE蛋白表达的影响。方法对哮喘患儿和健康儿童各50例,通过甲基化特异性聚合酶链反应(MSP)进行甲基化分析IL-4基因甲基化表达,酶联免疫吸附测定法检测患儿和健康儿童IL-4、IgE蛋白水平。结果哮喘患儿血清IL-4、IgE水平均明显高于健康对照组,组间比较差异均有统计学意义(P值均〈0.05);哮喘患儿IL-4与IgE水平呈正相关(r值分别为0.454、0.425,P〈0.01);哮喘患儿IL-4基因启动子区完全甲基化率56%(28/50),未甲基化率为14%(7/50),甲基化与未甲基化共存率为30%(15/50);健康儿童IL-4基因启动子区完全甲基化率为16%(8/50),未甲基化率24%(12/50),甲基化与未甲基化共存率为60%(30/50),哮喘患儿IL-4、INF-γ基因甲基化率与健康儿童比较差异均有统计学意义(P〈0.05)。结论哮喘患儿IL-4基因甲基化率高于健康儿童,IL-4、IgE蛋白表达与IL-4基因甲基化呈正相关;IL-4基因甲基化这一表观遗传现象在哮喘发病机制中有一定的作用。Objective To study the gene methylation in children with bronchial asthma (asthma) and IL 4 expression differences in healthy children and impact on IL-4, IgE protein expression. Methods Fifty cases of children with asthma and healthy children, by methylation specific PCR (MSP) methylation analysis IL-4 gene methylation expression, enzyme-linked immunosorbent assay to detect sick children and healthy chiidren IL-4, IgE protein levels. Results Children with asthma serum IL 4, IgE levels were significantly higher than that of healthy controls, comparison between groups were statistically significant (all P〈0.05). Children with asthma IL4 and IgE levels were positively correlated relationship, correlation coefficient r =0. 454 and r 0. 425 respectively ( P〈0.01). Children with asthma IL-4 gene promoter region methylation rate was 56% (28/50), completely not methylation rate was 14 % (7/50), methylation and methylation coexistence rate was 30% (15/50). Healthy children IL-4 gene promoter region methylation rate was 16% (8/50), completely not methylation rate was 24% (12/50), the coexistence of methylation and methylation rate was 60% (30/50), children with asthma IL-4, INF-gamma gene methylation rate compared with healthy children were statistically significant ( P〈0.05). Conclusions Children with asthma IL-4 gene methylation rate is higher than that of healthy children, IL-4, IgE protein expression and IL-4 gene methylation were positively correlated. IL-4 gene methylation this epigenetic phenomenon has a certain role in the pathogenesis of asthma.
