摘  要:目的研究白藜芦醇对大鼠肝癌细胞CBRH7919凋亡的诱导作用。方法将大鼠肝癌细胞CBRH7919分为2组:A组用80μmol·L^(-1)的白藜芦醇处理,B组加入等量的二甲基亚砜(DMSO);选取大鼠正常肝细胞BRL-3A作为对照组,也将细胞分为2组:C组用80μmol·L^(-1)的白藜芦醇处理,D组加入等量的DMSO;比较各组细胞的细胞活性、细胞周期变化、caspase-3、bcl-2和bax的表达水平和线粒体膜电位。结果 A组细胞的细胞活力较B组细胞的活力明显减弱(P<0.05)。A、B、C、D组细胞G0/G1分别为(35.67±3.38)%,(48.12±5.01)%,(41.98±4.05)%,(41.87±4.11)%。A组细胞处于G_0/G_1期细胞比例明显少于B组(P<0.05),且2组细胞的caspase-3、bcl-2和bax的表达水平和线粒体膜电位有明显差异(P<0.05)。C组和D组细胞的细胞活性、细胞周期变化、caspase-3、bcl-2和bax的表达水平和线粒体膜电位无明显差异(P>0.05)。结论白藜芦醇可有效诱导大鼠肝癌细胞CBRH7919的凋亡,其可能是通过调控Bcl-2/Bax的表达和引起线粒体膜的去极化发挥抗肿瘤作用。Objective To study the effects of resveratrol on apoptosis of hepatoma cells CBRH7919 in rat. Methods Rat hepatoma cells CBRH7919 were divided into 2 groups,group A was treated with 80μmol·L^-1 resveratrol,group B was treated with dimethyl sulfoxide( DMSO). At the same time,BRL- 3A cells were divided into group C( 80 μmol·L^-1 resveratrol) and group D( DMSO). Proliferation activity,cell cycle distribution,expression level of caspase- 3,bcl- 2 and bax and mitochondrial memberane potential were analyzed for comparison in 4groups. Results The proliferation activity of group A was less than that of group B,the difference had statistical significance( P〈0. 05). In group A,the ratio of G_0/G_1was( 35. 67 ± 3. 38) %,that of group B was( 48. 12 ±5. 01) %,that of group C was( 41. 98 ± 4. 05) %,and that of group D was( 41. 87 ± 4. 11) %. In group A,the ratio of cells in G0/G1 was higher than that of group B, the difference had statistical significance( P〈0. 05). The expression level of caspase- 3,bcl- 2 and bax and mitochondrial memberane potential had statistical significance had statistical difference between group A and group B( P〈0. 05). Proliferation activity,cell cycle distribution,expression level of caspase- 3,bcl- 2and bax and mitochondrial memberane potential had no statisticaldifference between group C and group D( P〉0. 05). Conclusion Resveratrol can induce the cell apoptosis of rat hepatoma cells CBRH7919,which is relative to regulating expression level of bcl- 2 / bax and depolarization of mitochondrial memberane potential.
