摘 要:为了研究家禽和人感染H7N9禽流感病毒血凝素及其表达特性的演变,试验从全球禽流感基因共享数据库(GISAID)和美国国家生物技术信息中心(Gen Bank)中获取我国家禽和人感染H7N9禽流感病毒血凝素(hemagglutinin,HA)核酸序列,用生物信息学软件Clustalx 2.0分析以家禽H7N9病毒为参照人感染毒株HA蛋白氨基酸变化和HA核苷酸同源性,并探讨其表达特性的演化。结果表明:2014年部分人感染毒株发生氨基酸替换,且病毒出现地方区域分支;2015年毒株均发生氨基酸替换且替换位点数量多于2014年,病毒保持区域分化;2014年江西家禽感染毒株与2014年、2015年多地人感染毒株具密切亲缘关系。说明氨基酸替换增多可能增强了H7N9禽流感病毒对人的感染力,病毒基因演变表达特性显现地域性特征,应高度关注江西家禽中H7N9病毒的演化趋势。To study the hemagglutinin of poultry and human infected with avian influenza H7N9 virus and the evolution of its expression characteristics, the nucleic acid sequences of hemagglutinin (HA) of China poultry and human infected with avian influenza H7N9 virus were obtained from GISAID and GenBank databases. The changes in the HA protein amino acids and HA nucleic acid homology in the H7N9 virus strains from human infected were analyzed using bioinformatics software Clustalx 2.0, taking avian influenza HTN9 virus from poultry as a reference, while the evolution of its expression characteristics was investigated. The results showed that amino acid substitutions occurred at partial strains of avian influenza HTN9 virus from human infected in 2014, and the virus appeared the regional branch ; the amino acid substitutions occurred at all strains in 2015, and the number of substitution sites in 2015 was more than in 2014, while the virus maintained regional diversification; the strains from poultry infected in Jiangxi province in 2014 had close kinship with the strains from human infected in many regions in 2014 and 2015. The results indicate that the increase of amino acid substitutions can enhance the infectivity of avian influenza HTN9 virus against human, and the viral gene shows geographical features due to the evolution of its expression characteristics, thus, it should be highly concerned about the evolutionary trend of avian influenza H7N9 virus in poultry from Jiangxi province.
