摘  要:为了研究ADAMTS-1基因的重要功能区与湖羊繁殖性状的关联性,试验采用湖羊作为研究对象,以我国固有品种小尾寒羊、洼地绵羊、阿勒泰羊作为参照群体,拟以ADAMTS-1去解链金属蛋白酶基因作为繁殖性状的候选基因,根据NCBI发表的ADAMTS-1基因序列设计引物,利用PCRSSCP方法检测4种绵羊ADAMTS-1基因第7,8外显子的多态性,分析绵羊ADAMTS-1基因部分重要功能区单核苷酸多态性及遗传变异水平。结果表明:ADAMTS-1第7外显子发现了1处A→G的碱基突变,除小尾寒羊外,其他3种羊的基因型分布均符合Hardy-Weinberg平衡,各基因型频率分布在4种绵羊间的差异均不显著;第8外显子发现了1处T→C的碱基突变,除湖羊外,其他3种绵羊的基因型分布全部符合Hardy-Weinberg平衡;第7外显子的GG基因型在产羔数上比GH基因型高0.094 7;第8外显子的IJ基因型在产羔数上比II基因型高0.053 3,说明GG和IJ基因型对湖羊的产羔性状有一定的影响,但是两个位点的不同基因型在产羔数上差异均未达显著水平(P>0.05)。说明各基因型与产羔性状关联性表现为差异不显著,初步认为ADAMTS-1基因不能作为湖羊高繁殖力的候选基因。To study the association between the important functional regions of ADAMTS - 1 gene and reproductive traits of Hu sheep, Hu sheep was used as research object, taking Chinese indigenous breeds as reference populations, which were small - tailed Han sheep, Wadi sheep, and Ahay sheep. A disintegrin and metalloproteinase with thrombospondin motifs ( ADAMTS - 1 ) gene was selected as a candidate gene for reproductive traits, and then the primers were designed according to the ADAMTS - 1 gene sequences published in NCBI. PCR - SSCP method was used to detect the polymorphisms of exon 7 and exon 8 of ADAMTS - 1 gene for four kinds of sheep, and then the single nucleotide polymorphisms and the levels of genetic variation in some important functional regions of the ADAMTS - 1 gene were analyzed. The result showed that A to G base mutation in the exon 7 was identified, and the genotype distributions conformed to Hardy - Weinberg equilibrium in four kinds of sheep except small - tailed Han sheep, and there were no significant differences in the distributions of genotype frequencies among four kinds of sheep. T to C mutation in the exon 8 was identified, and the genotype distributions conformed to Hardy - Weinberg equilibrium in four kinds of sheep except Hu sheep. GG genotype was 0. 094 7 higher than GH genotype on litter size in the exon 7, and IJ genotype was 0. 053 3 higher than II genotype on litter size in the exon 8, indicating that GG and IJ genotypes had certain effects on the lambing traits in Hu sheep. However, the differences of different genotypes on litter size at the two loci were not significant ( P 〉 0.05 ). The results indicate that there are no significant differences between genotypes and lambing traits. It is preliminary considered that ADAMTS - 1 gene can' t be a candidate genefor Hu sheep with high fecundity.
