摘 要:为了研究吉林省犊牛腹泻病病原,试验采用灭菌棉拭子无菌采集吉林省部分地区奶牛场发生典型腹泻症状的64份犊牛直肠拭子样品进行病原菌的分离、理化、16S r DNA鉴定,O因子血清型鉴定,致病性分析以及强毒力岛(HPI)核心区irp2、fyu A和ybt A基因携带情况检测。结果表明:试验共分离出致病性大肠杆菌39株,irp2基因阳性率85%(33/39),即33株大肠杆菌HPI阳性。HPI阳性分离株中检出fyu A基因10株,阳性率为30%(10/33);ybt A基因6株,阳性率为18%(6/33)。共有15株大肠杆菌分离株对小鼠有较高的致病性。39株菌共覆盖8种血清型,其中O14为优势血清型,占定型菌株的56%。说明HPI在犊牛腹泻大肠杆菌中广泛分布且HPI的携带率与菌株的致病性呈明显的正相关。To study the pathogen of calf diarrhea disease in Jilin Province, 64 rectal swab samples from calves with typical symptom of diarrhea were antiseptically collected in some dairy farms from partial areas of Jilin province using sterile cotton swabs. The samples were used for the isolation of pathogens, physical and chemical identification, 16s rDNA identification, identification of O serotypes, pathogenic analysis, as well as the detection of the irp2, fyuA and ybtA genes carried in the core region of HPI, respectively. The result showed that 39 pathogenic Escherichia coli strains were isolated. The positive rate of the irp2 gene was 85% (33/39), namely, there were 33 HPI -positive isolates of Escherichia coli. Ten isolates with the fyuA gene and 6 isolates with the ybtA gene were detected in the HPI - positive isolates, and their positive rates were 30% (10/33) and 18% (6/33), respectively. A total of 15 isolates of Escherichia eoli had high pathogenicity in mice. The 39 isolates covered 8 kinds of serotypes, thereinto, O14 was a dominant serotype, which accounted for 56% of stereotype strains. The results indicate that the HPI is widely distributed in Escherichia coli of calf diarrhea, and there is obviously positive correlation between the carrying rate of HPI and the pathogenicity of bacterial strain.
