摘  要:目的观察重组人白细胞介素-11(rhIL-11)对脓毒症相关肠道损伤的保护作用。方法将SD大鼠随机分为正常组(10只)、假手术组(对照组,10只)、脓毒症模型组(脓毒症组,30只)、治疗组(rhIL-11组,30只)。以盲肠结扎穿孔法(CLP)制作脓毒症大鼠模型,治疗组在脓毒症模型基础上给予500Ixg/(kg·d)rhIL-11皮下注射,共3d;造模成功后24、48h留取各组大鼠空肠标本进行病理学观察;采用RT—PCR法检测空肠组织内凋亡相关基因BaxmRNA的表达,Western blot法检测JAK1蛋白的表达。结果①病理形态学染色提示,治疗组大鼠空肠黏膜炎症、坏死明显少于脓毒症组;②rhIL-11能下调大鼠空肠组织内BaxmRNA的表达:治疗组大鼠空肠组织内BaxmRNA表达指数低于脓毒症组(P〈0.05);③rhIL-11能上调大鼠空肠组织内JAK1蛋白的表达:治疗组大鼠空肠组织内JAK1蛋白表达高于脓毒症组(P〈0.05)。结论rhIL-11可能通过激活JAK1/STAT3信号通路提高肠道JAK1蛋白的表达、降低BaxmRNA的表达而减轻脓毒症相关的小肠损伤。Objective To evaluate the protective effect of recombinant human interleukin - 11 (rhIL-11 ) on intestinal injury associated with sepsis. Methods Eighty SD rats were divided into rhIL- 11 treatment group (30 rats), sepsis group (30 rats), normal group (10 rats) and sham group ( 10 rats) randomly. Septic rat model was established by cecal ligation and perforation (CLP). rhIL-11 treatment group and sham group were given subcutaneous injection of rhIL-11 500 μg/(kg·d) additionally for 3 days. However, rats of sepsis group or normal group were injected with balanced solution ( 10 mL/kg, d). Samples of intestine were collected at 24 h and 48 h after CLP to perform histopathological study and molecular analysis. The mRNA expressions of Bax were evaluated by transcription-polymerase chain reaction. The protein expressions of JAK1 in lung were assayed by western blot. Results (1)The injury and the necrosis of small intestinal mucosal were reduced significantly in treatment group compared with that of sepsis group. (2)The expression level of Bax mRNA was lower in treatment group compared with that of sepsis group (P 〈 0.05). (3)The level of JAK1 protein expression in the treatment group was observably higher than that of the sepsis, sham, and normal groups (P 〈 0.05). Conclusion rhIL- 11 can reduce the intestinal injury associated with sepsis, the main mechanism maybe rhIL- 11 can regulate JAK/STAT signal pathway and reduce the expression of Bax.
