摘 要:为优化蝴蝶兰类原球茎诱导和增殖再生条件,以蝴蝶兰品种‘红太阳’无菌播种形成的原球茎为材料,研究了取材时间、植物生长调节剂(6-BA、NAA)、碳源等对类原球茎诱导和增殖及分化再生的影响,结果表明,生长45d的蝴蝶兰原球茎最适于类原球茎诱导和增殖培养。最佳的类原球茎诱导和增殖培养基为:花宝1号3.5g/L+6-BA 5.0mg/L+NAA 0.7mg/L+白糖20g/L+琼脂6.0g/L+活性炭2.0g/L,pH为5.4-5.8,最高增殖系数可以达到17.33,比已报道的最高增殖系数提高了35.39%;类原球茎最佳分化培养基为:花宝1号3.5g/L+6-BA 3.0mg/L+NAA 0.5mg/L+白糖20g/L+琼脂6.0g/L+活性炭2.0g/L,pH为5.4-5.8,最高分化率可达96.67%,生根率最高可达到92%。In order to establish multiplication and regeneration system of protocorm-like bodies in Phalaenopsis.The effects of different growth days after germination,different concentrations of plant growth regulator in medium,different carbon on protocorm-like bodies induction and proliferation and regeneration efficiency of different hormone concentrations were studied in Phalaenopsis Var.Red.Sun.This experiment suggests that the high efficiency and economical protocorm multiplication system is Hyponex 3.5g/L+6-BA 5.0mg/L+NAA 0.7mg/L+sugar 20g/L+agar 6.0g/L+activated carbon 2.0g/L,pH 5.4-5.8,using protocorm grew to 45 th day after germination.The maximum multiplication rate can be reached to 17.33.It has increased 35.39%compared with the previous reports.The optimal regeneration system is that Hyponex 3.5g/L+6-BA 3.0mg/L+NAA 0.5mg/L+sugar 20g/L+agar 6.0g/L+activated carbon 2.0g/L,pH 5.4-5.8,regeneration rate can be reached to 96.67%,rooting rate can be reached to92%,and seedlings grow well.
