摘  要:采集阳泉新景煤矿矸石山及其下游土壤样品,考察其浸出液对人类肝癌细胞系HepG2和赤子爱胜蚓的生物毒性作用,并对矸石堆放区域重金属污染现状及潜在生态风险进行评价。研究发现,矸石山及其周边土壤浸出液明显抑制HepG2细胞活性;随距矸石山距离的增加,土壤浸出液造成细胞存活率的距离依赖性下降,但即使下游100m村庄附近土壤浸出液对细胞活性的抑制作用也极为显著,细胞存活率为对照组的40%(P<0.001)。在此基础上,考察不同浓度矸石及其下游村庄附近土壤浸出液对HepG2细胞活性和赤子爱胜蚓脂质过氧化产物丙二醛(MDA)含量的影响。结果表明,矸石及其下游村庄土样均抑制HepG2细胞活性并诱导蚯蚓MDA水平升高,呈现明显的剂量-效应关系。即在剂量浓度为1∶1时,村庄附近100m土样和矸石浸出液对HepG2细胞活性的抑制作用达到最大,细胞存活率分别是对照组的28%(P<0.001),和56%(P<0.001);同时诱导赤子爱胜蚓MDA水平达到最高,分别为对照组的1.36倍(P<0.001)和1.23倍(P<0.05)。实验结果表明,即使下游100m村庄附近土壤浸出液仍表现出与矸石浸出液相当甚至更高的的生物毒性效应。采用单因子指数法、内梅罗综合污染指数法和潜在生态危害指数法分析评价矸石山堆放区域重金属污染状况,结果表明,矸石山下游650m以内的村庄及农田土壤均达到重度污染,以Cd污染为主。这一结果提示,矸石堆放会导致周边土壤生态风险。We collected the coal gangue and the downstream soil samples from Xinjing coal mine in Yangquan,and investigated the biological toxicity effects of the soil leachates on human hepatocellular carcinoma cell line HepG2 and the Eisenia Foetida.In addition,we evaluated the current status and potential ecological risk of the heavy metal pollution of coal gangue heap area.The results indicated that the leachates of the coal gangue and the surrounding soils significantly inhibited the cell viability of HepG2,and with the increase distances from gangue dump to downstream sampling sites,the soil leachates cause the distancedependent reduction of cell survival rate.The soil leachate nearing the village,extracted from the downstream 100 msoil sample of coal gangue,exhibited a significant biological toxicity effect to the cell viability,which has reduced to 40%(P〈0.001)of the control.Moreover,we determined the effects of coal gangue and its downstream village soil leachates on the cell viability of HepG2 and the lipid peroxidation product malondialdehyde(MDA)content of Eisenia Foetida.The results showed that the leachates of coal gangue and its downstream village soil inhibited cell viability of HepG2 and increased MDA content of Eisenia foetida,and presented an obvious concentration-effect relationship.In the dose concentration of 1∶1,the inhibitory effects of the leachates from the village 100 msoil and gangue on HepG2 cells were significantly increased,and the cell viability decreased to 28%(P〈0.001)and 56%(P〈0.001)of the control,respectively.And the MDA level of E.foetida has been induced,which increased to 1.36times(P〈0.001)and 1.23times(P〈0.05)of the control,respectively.The results showed that the downstream 100 m soil samples of coal gangue nearing the village is comparable to the leachate of gangue,even caused more significant biotoxicity than gangue.We evaluated the current status of the heavy metal pollution of coal gangue heap area using single factor index method,Nemerow compr
