摘  要:目的探讨不同程度的噪声性听力损伤(NIHL)与大鼠耳蜗螺旋神经节细胞(SGC)Caspase-3表达水平的关系,分析SGC噪声损伤的凋亡机制。方法 SPF级雄性SD大鼠30只随机分为3组,每组10只。除对照组外,其余2组分别予以90、110 d B SPL的白噪声暴露,每天6 h,连续28 d,暴露前1 d和暴露结束后第7 d进行听性脑干反应(ABR)检测和SGC病理学检查,免疫组化法分析SGC的Caspase-3表达水平。结果各组间大鼠平均永久性听阈位移(PTS)水平变化差异有统计学意义(F=1 103.21,P<0.01),随噪声暴露强度的增加,PTS水平也增加;各组大鼠SGC的Caspase-3平均灰度值(IA)差异有统计学意义(F=1 190.93,P<0.01),随噪声暴露强度的增加,IA值亦增加;病理学观察发现各噪声暴露组均出现SGC的损伤,110 d B SPL噪声暴露组出现明显的SGC溶解现象。结论轻度NIHL即可诱发耳蜗SGC的凋亡。[Objective]To investigate the relationship between different degree of noise-induced hearing loss(NIHL) and expression level of Caspase-3 in cochlear spiral ganglion cells(SGC) of rats,analyze the apoptosis mechanism of SGC noise damage.[Methods]30 SPF male rats were randomly divided into three groups,10 rats in each group.Except the control group,two exposed groups were given the white noise exposure of 90 and 110 d B SPL respectively,6 hours a day for 28 days.Auditory brainstem response(ABR)test and SGC pathological examination were performed at the day before exposure and 7 th day after exposure.The expression level of Caspase-3 in SGC was analyzed by immunohistochemistry method.[Results]The difference in level of average permanent threshold shift(PTS) was statistically significant among three groups(F=1 103.21,P〈.01),PTS level increased with increasing of noise intensity.There was statistically significant difference in average gray value(IA) of Caspase-3 among three groups(F =1 190.93,P 0.01),IA value increased with increasing of noise intensity.The pathological observation found that there was SCG damage in two exposed groups,and dissolution of SCG was obviously observed in exposed groups with 110 d B SPL.[Conclusion]Mild NIHL can induce the apoptosis of cochlear SGC.
