摘　　要：为筛选出具有较高除草活性的微生物菌株,并对其次级代谢产物中除草活性物质进行分离纯化,本试验从西瓜细菌性果腐病病叶上分离纯化微生物菌株,利用茎叶处理法测定了其次级代谢产物除草活性,利用16S rDNA序列和生理生化试验对除草活性最高的菌株进行了种属鉴定,进而利用硅胶柱层析结合高效液相色谱(HPLC)法对其次级代谢产物中的除草活性物质进行了分离纯化。结果表明:共分离得到4株具有除草活性的细菌,其中菌株XG1对马唐的除草活性最高,将其鉴定为蜡样芽孢杆菌(Bacillus cereus)。发酵液经液-液萃取,硅胶柱层析和HPLC分离纯化得到了保留时间为15.757 min的除草活性物质Ⅰ。本研究证明,菌株XG1具有开发成微生物源除草剂的潜力,并为今后分离纯化细菌次级代谢产物的除草活性物质和开发新型细菌除草剂奠定了基础。For screening out microbial strains with high herbicidal activity and purifying active substances produced by the strain metabolites,our test isolated microbial strains from the watermelon leaves infected by watermelon bacterial fruit rot,using cauline leaf daub method to measure the herbicidal activity of the strains＇ second metabolite.The 16 SrDNA sequence and physicochemical tests were used to identify the strain having the highest herbicidal activity,and the second metabolites were isolated and purified by silica gel column chromatographycombined with high performance liquid chromatography（HPLC）method.The results indicated that 4bacterial strains with herbicidal activity were isolated,and the strain XG1 had the highest activity on Digitaria sanguinalis,so it was identified as Bacillus cereus.The substanceⅠwas isolated from fermented liquid by liquid-liquid extraction,silica gel column chromatography and HPLC purification,with the retention time of 15.757 min.The research concluded that the strain XG1 had the potential to be developed as a microbe-herbicide,and the experiment laid a foundation for purifying active substances produced by the second metabolites and developing a new bacterial herbicide.
