摘 要:以‘鲲旱2号’成熟胚为外植体,通过农杆菌介导法将抗虫基因PPA导入旱稻愈伤组织中,以期获得抗虫转基因旱稻植株。对影响愈伤组织诱导效果的2,4-D浓度进行了研究。结果表明:当2,4-D浓度为2mg/L时,愈伤组织诱导效果最佳。对获得的转化植株进行PCR检测证明PPA基因已成功整合到旱稻基因组DNA。对阳性植株的后代喷施0.8mg/mL除草剂检测表明,转基因植株后代具有良好的除草剂抗性。In order to get the insect-resistant transgenic upland rice plants,the mature embryos of upland rice were used as explants,and the insect-resistant gene PPA was transformed into the upland rice callus by Agrobacterium-mediated method.The concentration of 2,4-D influencing the callus induction was studied.The results showed that when the 2,4-D concentration was 2mg/L,the effect of callus induction was the best.The PPAgene was proved to be transformed into the genomic DNA of upland rice plants by PCR test.The later generations of the PCR positive plants and wild type plants were sprayed with 0.8mg/mL weedicide Basta solution,and the result indicated that the transgenic plants gained high tolerance to the herbicide.
