摘 要:目的:分析并鉴别桑叶水提取物化学成分及其口服后大鼠血清中的成分。方法:采用HPLC对桑叶水提物及灌胃后大鼠血清样品进行分离,用静电场轨道阱组合式高分辨质谱仪进行在线离子检测、二级碎片离子扫描,分析鉴别桑叶水提物体外成分及血中代谢成分。结果:桑叶水提物中初步鉴定了11个化学成分,分别为异绿原酸(1),5,7-二羟基香豆素-7-O-β-D-吡喃葡萄糖苷(2),东莨菪苷(3),绿原酸(4),山柰酚-3,7-二-O-D-吡喃糖苷(5),4-咖啡酰奎宁酸甲酯(6),芦丁(7),金丝桃苷(8),异槲皮苷(9),紫云英苷(10),异鼠李素-3-O-葡萄糖苷(11)。初步鉴定血中成分30个,多为上述苷类的苷元代谢产物,血清中未发现上述11个原型成分。结论:该方法可成功用于桑叶水提物及其口服后血中成分的鉴别,为桑叶进一步的药效研究及二次开发提供了参考。Objective: To analyze and identify the chemical compounds in Mori Folium aqueous extract and the components in serum of rats after oral administration. Method: HPLC was employed to analyze the compounds in Mori Folium aqueous extract as well as in rat serum after intragastric administration. LTQ-Orbitrap XL high-resolution mass spectrometer was used for online ions detection and secondary ions scanning to identify the compounds in Mori Folium aqueous extract and its metabolites in blood samples. Result: The 11 chemical compounds were identified in the Mori Folium aqueous extract, including isochlorogenic acid( 1), 5, 7-dlhydroxycommarin-7-O-β-D-glucopyranoside( 2), scopolin( 3), chlorogenic acid( 4), kaempferol-3,7-dlO-β-D-glucopyranoside( 5), 4-caffeoylquinic acid methyl ester( 6), rutin( 7), hyperoside( 8),isoquercitrin( 9), astragalin( 10), and isorhamnetin-3-O-glucoside( 11). The 30 compounds were preliminarily identified in the blood, and major of, them were aglycone metabolites. The above 11 original compounds were not found in serum. Conclusion: The present method can be used for the identification of the compounds in Mori Folium aqueous extract and blood of rats after oral administration, providing reference for further efficacy study and secondary development of Mori Folium.
