摘  要:目的:分别采用水蒸气蒸馏法(SD)和超临界CO2萃取法(SFE)提取回族药木香油方中挥发油和小极性成分,利用气质联用法鉴定分析各成分及相对含量,比较不同提取方法所得成分种类的差异。方法:Shimadzu QP2010 plus GC-MS条件:Rxi-5Sil MS石英毛细管柱(0.25 mm×30 m,0.25μm),起始温度50℃,保留1 min,以8℃·min^(-1)升温至150℃维持3min,以3℃·min^(-1)升温至210℃维持3 min,以15℃·min^(-1)升温至280℃维持15 min至完成分析;载气氦气,柱流量1.33 m L·min^(-1),分流比25∶1,进样口温度250℃,EI电离源70 e V,离子源温度230℃,扫描范围m/z 35~500。结果:SD法提取挥发油得率为0.8%,鉴定出25个化合物,占挥发油总峰面积的96.28%;SFE法提取挥发油得率为2.91%,共鉴定出36个化合物,占成分总峰面积的97.98%;结论:采用SD法和SFE法提取得到的回族药香药木香油方中小极性成分在种类上有较大差异,GC-MS可用于鉴定和测定香药小极性成分,为下一步阐明回族药香药的活性物质基础提供了试验数据。Objective: To extract the volatile oil and small polar components of the Hui formula Muxiang oil by steam distillation( SD) and supercritical CO2 fluid extraction( SFE),respectively; analyze and identify the components and their contents with Gas Chromatography-Mass Spectromet( GC-MS),and compare the differences in types of components between different extraction methods. Method: The mass spectrometric conditions were set as follows: Shimadzu QP2010 plus GC-MS,Rxi-5Sil MS quartz capillary column( 0. 25 mm × 30 m,0. 25 μm),the starting temperature of 50 ℃,maintained for 1 min,heated to 150 ℃ with 8 ℃·min-1,maintained for 3 min,then heated to 210 ℃ with 3 ℃·min-1,maintained for 3 min,and then heated to 280 ℃ with 15 ℃·min-1,maintained for 15 min until completion of analysis; helium as the carrier gas,column flow rate at 1. 33 m L·min^(-1),split ratio of25∶ 1,inlet temperature of 250 ℃,EI ionization source of 70 e V,ion source temperature of 230 ℃,and scan range of m / z 35-500. Result: Yield of volatile oil was 0. 8% and 2. 91% respectively in SD and SFE methods; 25 compounds were identified, accounting for 96. 28% of the total peak area of volatile oil in SD method; 36 compounds were identified,accounting for 97. 98% of the total peak area of volatile oil in SFE method. Conclusion:There was significant difference in types of components contained in the volatile oil between SD and SFE methods,and GC-MS could be used to identify the small polar components of the medicine,providing experimental basis for further development and utilization of Hui medicine.
