摘  要:从郑州市纳污河流贾鲁河中筛选出一株能高效去除氨氮的细菌,并对其生长特性进行研究。采用传统的微生物分离纯化的方法对所采集的样品进行筛选,用分子生物学的方法对菌株进行鉴定,并通过单因素实验对所选菌株的培养条件进行初步优化。结果显示,所筛选菌株为假黄单胞菌属(Pseudoxanthomonas sp.),命名为C2。该菌株能在有机物存在的条件下进行异养硝化作用,并快速繁殖,当接种量为10%时,培养4 h后即进入对数生长期。该菌株异养硝化的最佳接种量为3%,最适碳源为丁二酸钠,最适p H为6-9,适宜的温度范围为25-30℃,摇床转速对氨氮的去除效果影响不大。菌株C2对氨氮的去除效果较好,对于河水中氨氮的去除具有一定的应用价值。The objective of this study is to isolate a strain of heterotrophic nitrification bacterium with high efficiency of removing ammonia-nitrogen from Jialu River of colleeting wastewater at Zhengzhou City of Henan Province, and to study its growth characteristics. The collected samples were screened by the traditional methods of microorganism separation and purification, the strain was identified using molecular biology methods, and the culture conditions of the strain were optimized by single factor experiment. Based on 16S rDNA sequence analysis, it was identified as Pseudoxanthomonas sp., designated as C2. The strain C2 had heterotrophic nitrification and grew rapidly while organic substances existed. C2 entered the logarithmic phase after 4 hours of culturing when the inoculation quantity was 10%. The optimal condition for heterotrophic nitrification was as follows : the inoculation quantity was 3%, the sodium succinate was taken as the only carbon source, the initial pH was 6-9, and the incubation temperature ranged from 25-30"C ; the shaking speed had little effect on the activity of removing ammonia-nitrogen in this experiment. In conclusion, the removal rate of ammonia-nitrogen is relatively high, which demonstrates that it is promising to be applied in removing ammonia-nitrogen from practical water bodies.
