摘  要:为研究樱桃谷雏鸭感染坦布苏病毒后的潜伏期、排毒规律及病毒在血液、组织中载量的变化,本试验将110只10日龄健康樱桃谷雏鸭分为3组,其中,静脉接种组70只,点眼滴鼻组及自然感染组各20只。经静脉、点眼滴鼻分别接种坦布苏病毒SDLY株,每只3.5mL病毒尿囊液(3.11×10^(-3) EID_(50)/只),自然感染组不接种病毒,与静脉接种组混养。接种后,每隔2d采集泄殖腔棉试子和血液,并随机剖杀3只,应用建立的荧光定量PCR检测病毒的排毒规律及在血液、组织中载量的变化。结果显示,3个组的樱桃谷雏鸭感染坦布苏病毒的潜伏期均一致,平均为24h;樱桃谷雏鸭在接种病毒后的1~25d均可排毒,感染后5d出现排毒高峰,其次为11、17d出现2个小排毒高峰,自然感染组出现高峰的时间比静脉及点眼滴鼻组晚2d;静脉接种组病毒在血液内的载量较其他2组明显低,高峰出现在感染后7、19d,点眼滴鼻组高峰同静脉接种组一致,出现在感染后7、19d,自然感染组血液中的病毒载量高峰出现在感染后9、21d;组织中病毒载量较高的器官有脑、胸腺、法氏囊、胰腺、心脏,其均在感染后5d出现先升高后又降低,并在13d左右升至高峰。研究表明,坦布苏病毒对樱桃谷雏鸭的感染能力较强,潜伏期短,病毒可通过泄殖腔向外界排毒,排毒时间较长,病毒随血液循环迅速侵入各组织器官。To investigate the period of incubation and dynamics of viral shedding and the detection in blood and diverse tissues of Cherry Valley ducklings infected with Tembusu virus,110 ducklings were inoculated with the duck-origin Tembusu virus(TMUV)SDLY strain each duckling3.5mL virus allantoic fluid intravenously or intranasally at 10-day-old.The ducklings were divided into groups of three:70ducklings for intravenous injection group,20 ducklings for intraocular-nasal injection group and 20 ducklings for naturally infected group.The infected naturally group was fedwith intravenous injection ducklings,without any other treatment.After the ducklings infected the TMUV,cloaca swabs and blood samples were collected and 3ducklings were killed at random every two days.We recorded both the clinical signs and the incubation period of the ducklings which have infected the TMUV.Furthermore,SYBR GreenⅠfluorescence quantitative RT-PCR analysis of viral shedding and detection in blood and diverse tissues were also performed.The results show that the incubation period of 3groups ducklings is consistent,average 24 h.Viral RNA can detected in cloacal swabs from the first day to 25 th day and the content of Tembusu virus in cloacal swabs is higher on the 5th,11 th and 17 th day,and reached a peak on the 5th day,were two days late in ducklings infected naturally than other two groups.The detection of Tembusu virus in blood of the intravenous injection group is significantly lower than the other two groups,and reached a peak on the 7th and 19 th day,the group intraocular-nasal injection also reached a peak on the 7th and 19 th day.The content of Tembusu virus in blood reached The highst value on 9th and 21 th day in ducklings infected naturally.Higher viral copy numbers in the tissues,especially in vital organs such as the brain,thymus,bursa of Fabricius,pancreas and the heart,firstly increased at the 5th day and reduced then,finally climbed to the highest at day 13.The experiment results indicated that Tembusu virus has a very st
