血府逐瘀汤对血凝素样氧化低密度脂蛋白受体-1等炎症因子表达的影响  被引量:6

摘  要:目的:研究血府逐瘀汤含药血清对血管内皮细胞血凝素样氧化低密度脂蛋白受体-1(LOX-1)、肿瘤坏死因子-α(TNF-α)、血管细胞黏附因子-1(VCAM-1)及细胞间黏附分子-1(ICAM-1)表达的影响,探讨血府逐瘀汤防治动脉粥样硬化的机制。方法:血管内皮细胞随机分为对照组、模型组、西药对照组和血府逐瘀汤组,用LPS刺激2 h后分别加入阿托伐他汀和血府逐瘀汤含药血清干预,24 h后收集细胞,用荧光定量PCR方法和Western blot方法测定LOX-1、TNF-α、VCAM-1及ICAM-1的基因及蛋白表达。结果:用LPS刺激内皮细胞后,引起LOX-1、TNF-α、VCAM-1及ICAM-1的表达升高,差异有统计学意义(与对照组比较P<0.01),而用血府逐瘀汤含药血清干预以后显著抑制LOX-1、TNF-α、VCAM-1及ICAM-1的表达,差异有统计学意义(与模型组比较P<0.05)。结论:血府逐瘀汤可抑制LOX-1、TNF-α、VCAM-1及ICAM-1等炎症因子的表达,这可能是其防治动脉粥样硬化作用的机制之一。Objective: To investigate the influence of serum containing Xuefu Zhuyu Decoction( XZD) on the expressions of LOX- 1,TNF- α,ICAM- 1 and VCAM- 1 in human vein endothelial cells( HUVECs) and to study its possible anti- atherosclerotic mechanism. Methods: HUVECs were cultured in vitro and equally divided into 4 groups in random: the normal control group,the model group,the western medicine group and the XZD group. HUVECs were stimulated with LPS for 2 h,and then treated separately with the drug- serum for 24 h. Finally,we measured the expressions of LOX- 1,TNF- α,ICAM- 1 and VCAM- 1 mRNA with real- time PCR,using western blotting method to analyze the expression of protein. Results: Protein and mRNA expressions of LOX- 1,TNF- α,ICAM- 1 and VCAM- 1 increased significantly after LPS stimulation( vs normal control group,P〈0. 01). Xuefu Zhuyu Decoction decreased the high expressions of LOX- 1,TNF- α,ICAM- 1 and VCAM- 1( vs model group,P〈0. 05). Conclusion: Xuefu Zhuyu Decoction can block the high expressions of LOX- 1,TNF- α,ICAM- 1 and VCAM- 1. Maybe it's the mechanism of Xuefu Zhuyu Decoction exerting the function of anti- αrtherosclerosis.
