摘  要:目的:首次引入ABCG2尿酸转运蛋白,探讨其在痛风性关节炎中的作用机制和穿山龙总皂苷的干预作用。方法:通过Real-time PCR和Western blot方法,检测高尿酸血症小鼠肾脏ABCG2转运蛋白的mRNA和蛋白表达水平。结果:模型组m ABCG2的mRNA和蛋白表达量均明显降低(P<0.001)。穿山龙总皂苷低剂量组可明显回调该基因的mRNA表达(P<0.001),穿山龙总皂苷高、中剂量组均可回调ABCG2 mRNA表达(P<0.05,P<0.001)。穿山龙总皂苷高、中、低剂量组均可回调ABCG2蛋白的表达(P<0.001,P<0.05和P<0.05)。结论:穿山龙总皂苷可通过对肾脏尿酸转运体ABCG2的调控来促进尿酸的排泄实现对高尿酸血症的降尿酸作用。Objective: First to introduce ABCG2 uric acid transporter,aiming at investigating its mechanism and whether the effects of total saponins from Rhizoma Dioscoreae Nipponicae on hyperuricemia mice induced by potassium oxonate in the current study. Methods: The mRNA and protein levels of ABCG2 uric acid transporter were analyzed by Real- time PCR and Western blotting methods. Results: The amount of mRNA and protein expression of m ABCG2 were significantly lowered in model group( P〈0. 001). The mRNA expression was reversed obviously in low- dose group( P〈0. 001) and the ABCG2 mRNA expression also reversed in high and middle dose groups( P〈0. 05,P〈0. 001).The high,middle and low dose groups' total saponins from Rhizoma Dioscoreae Nipponicae can reverse ABCG2 protein expression( P〈0. 001,P〈0. 05 and P〈0. 05). Conclusion: Total saponins from Rhizoma Dioscoreae Nipponicae could effectively reverse potassium oxonate- induced alterations in ABCG2 uric acid transporter mRNA and protein levels,resulting in enhancement of renal urate excretion in mice.
