摘 要:目的:探讨复方楂金颗粒(CZJG)中试产品对高脂饲料诱导的非酒精性脂肪肝炎大鼠脂质代谢及肝细胞超微结构的影响。方法:SD大鼠60只,随机分为正常组、模型组、易善复组(61.6 mg/kg)及复方楂金颗粒高、中、低剂量组(3.50、1.75、0.88 g/kg),每组10只。以高脂饲料喂养造模,给药12周后,处死所有大鼠,测定大鼠肝组织匀浆上清液TC、TG、FFA的含量;ELISA法检测大鼠肝匀浆中SOD、MDA、GSH-Px、TAOC的活性;透射电子显微镜观察肝细胞的超微结构。结果:与模型组比较,CZJG各剂量组大鼠肝组织匀浆TC含量均显著降低(P<0.05),中剂量组TG含量、高剂量组FFA含量显著降低(P<0.05);低剂量组SOD水平显著升高(P<0.01),高、低剂量组MDA水平显著降低(P<0.05;P<0.01)。电镜下可见模型组肝细胞超微结构异常,肝细胞胞浆脂滴聚积,线粒体嵴变少缩短,细胞核部分呈现固缩;CZJG高剂量组肝细胞胞浆可见大量脂滴,线粒体多数嵴清晰可见,细胞核呈圆形,无明显异常。结论:CZJG中试产品可有效调节NASH实验大鼠肝脏脂质代谢及抗脂质过氧化水平,其作用可能与改善线粒体代谢等有关。Objective: To investigate the effect of the trial product of Compound Zhajin Granules( CZJG) on lipid metabolism and liver cell ultrastructure in rats with non- alcoholic steatohepatitis( NASH) induced by high- fat diet.Methods: A total of 60 SD rats were randomly divided into control group,model group,Essentiale group( 61. 6 mg/kg)and CZJG high dose,medium dose and low dose groups( 3. 50 g/kg,1. 75 g/kg,0. 88 g/kg). Except the control group which was raised by standard animal feeds,others were all raised by high fat feeds to make NASH. At the end of 12 th week,all rats were sacrificed. The contents of TC,TG and FFA in liver homogenate supernatant were determined. ELISA was used to detect the activities of SOD,MDA,GSH- PX and TAOC. The ultrastructure of hepatocyte was observed by using transmission electron microscope. Results: Compared with the model group,the level of TC in all CZJG doses groups decreased significantly( P〈0. 05) and the level of TG in medium dose group and FFA in high dose group decreased significantly( P〈0. 05). The activity of SOD in low dose group significantly increased( P〈0. 05,P〈0. 01) and the activities of MDA in high and low dose groups were statistically significant( P〈0. 01). Electron microscope showed that the liver cell ultrastructure in model group was abnormal: accumulation of lipid droplets,reduction and disappearance of mitochondrial cristae and shrinkage and condensation of cell nuclei. In CZJG high dose group,lipid droplets were seen in the cytoplasm of liver cells and most mitochondrial cristae visible and nuclei round with no obvious abnormalities. Conclusion: The trial product of CZJG can effectively regulate the lipid metabolism and lipid peroxidation levels of experimental rats with NASH,which may be related to improving mitochondrial metabolism.
