从氧化应激谈白藜芦醇对高尿酸介导的非酒精性脂肪肝大鼠的影响

摘　　要：目的:探讨白藜芦醇对高尿酸介导的大鼠非酒精性脂肪肝氧化应激的影响。方法:雄性SD大鼠,给予改良的高酵母高脂饲料喂养合并氧嗪酸皮下注射建立高尿酸介导的非酒精性脂肪肝大鼠模型。36只SPF级雄性SD大鼠,随机分为3组:正常对照组(A组),模型组(B组),白藜芦醇治疗组(C组),每组12只。分别观察白藜芦醇对高尿酸介导的非酒精性脂肪肝大鼠体重、肝湿重、血清尿酸(UA)、超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX)含量的影响,并与模型组比较分析其病理学改变。结果:白藜芦醇治疗后,高尿酸介导的脂肪肝大鼠肝湿重均明显低于模型组(P<0.05),血清UA、MDA含量均明显降低(P<0.01),而血清SOD、GSH-PX含量均明显升高(P<0.01)。且白藜芦醇治疗后,肝组织脂肪变性及炎性反应较模型组减轻。结论:白藜芦醇可抑制高尿酸介导的大鼠非酒精性脂肪肝肝组织氧化应激,一定程度上对其具有治疗作用。Objective： To investigate the effect of resveratrol on the oxidative stress of nonalcoholic fatty liver disease rats with the high uric acid. Methods： Male SD rats were treated with modified high yeast and high fat diet and subcutaneous injection of oxonic acid potassium salt to build the model of NAFLD with high uric acid. A total of 36 SPF male SD rats were randomly divided into 3 groups： control group（ Group A）,model group（ Group B）,treatment group（ Group C）,12 rats in each group. We respectively observed body mass,liver wet weight,UA,SOD,MDA and GSH- PX in each group. And compared with the model group,the pathological changes were analyzed. Results： After resveratrol treatment,the liver wet weight of the rats was significantly lower than that of the model group（ P〈0. 05）. UA and MDA were significantly decreased（ P〈0. 01）,but SOD and GSH- PX significantly elevated（ P〈0. 01）. Besides,after resveratrol treatment,the range of fatty degeneration and inflammatory reaction in the liver tissue were smaller compared with those of the model group. Conclusion： Resveratrol can inhibit the oxidative stress. To some certain extent,it has therapeutic effect on the nonalcoholic fatty liver disease rats with high uric acid.
