三棱莪术组方对人胃癌细胞SGC-7901移植瘤裸鼠血清COX-2、VEGF和bFGF含量的影响

摘　　要：目的:研究三棱莪术组方对人胃癌细胞SGC-7901移植瘤裸鼠的抗肿瘤效应及对血清COX-2、VEGF和b FGF含量的影响,探讨其抑制胃癌的作用机制。方法:采用人胃癌细胞SGC-7901单细胞悬液接种方法建立裸鼠胃癌模型,三棱莪术组方灌胃给药(60 g/kg、30 g/kg、15 g/kg),阳性对照药氟尿嘧啶组腹腔注射给药(10 mg/kg),模型组灌服等量生理盐水。氟尿嘧啶组隔天腹腔注射给药1次,其余各组每天灌胃给药1次。裸鼠取瘤体,测量其体积并称重,取血,检测血清中COX-2、VEGF和b FGF含量。结果:与模型组比较,三棱莪术组方高、中、低剂量组裸鼠的肿瘤重量比模型组明显减轻,肿瘤生长受到抑制,抑瘤率分别为63.60%、61.52%、42.51%,具有显著性差异(P<0.05,P<0.01),抑瘤率与药物的浓度呈正相关。三棱莪术组方高、中、低剂量均可降低人胃癌SGC-7901细胞移植瘤裸鼠血清COX-2含量(P<0.05,P<0.01),三棱莪术组方高、中剂量可降低胃癌SGC-7901细胞移植瘤裸鼠血清VEGF和b FGF水平(P<0.05,P<0.01),并具有明显的剂量依赖性。结论:三棱莪术组方具有抑制人胃癌细胞SGC-7901移植瘤生长的作用,三棱莪术组方可通过降低人胃癌细胞SGC-7901移植瘤裸鼠血清COX-2、VEGF和b FGF水平从而达到抑制肿瘤生长的作用。Objective： To observe the anti- tumor activities and mechanisms of Sanleng Ezhu Zufang on BALB/c mice model of SGC- 7901 gastric carcinoma（ GC） and test the related proteins COX- 2,VEGF and b FGF in serum of miceby ELISA. Methods： In this study,nude mice were subcutaneously transplanted with SGC- 7901 gastric cancer cell line.When the tumor model formed,the mice were divided into Sanleng Ezhu Zufang high,medium and low dose groups（ 60 g/kg,30 g/kg,15 g/kg）,5- Fu（ Fluorouracil） group（ 10 mg/kg） and control group. Mice were treated for 8 weeks. After treatment,weight and volume of tissue tumor were tested as well as related proteins COX- 2,VEGF and b FGF were quantified in serum of mice model by ELISA. Results： The weight and volume of Sanleng Ezhu Zufang high,medium and low dose groups were reduced compared with those of the model group（ P〈0. 05,P〈0. 01） and inhibitory rates were63. 60%,61. 52% and 42. 51% respectively. The COX- 2 concentration in serum of Sanleng Ezhu Zufang high,medium and low dose groups were decreased（ P〈0. 05,P〈0. 01） and the VEGF and b FGF in serum of Sanleng Ezhu Zufang high and medium dose groups were decreased（ P〈0. 05,P〈0. 01）. Conclusion： The results revealed that Sanleng Ezhu Zufang can inhibit tumor growth by down- regulating the concentrations of COX- 2,VEGF and b FGF in serum.
