摘 要:目的研究血虚肝郁证大鼠海马CA1区磷酸二酯酶(PDE)不同亚型蛋白表达的变化及芍药苷的调控作用。方法分为空白组、模型组、芍药苷低剂量组(20 mg/kg)、芍药苷高剂量组(40mg/kg),造模第1天开始给药,空白组、模型组给等量纯水,共造模21 d。治疗结束,灌注固定后取脑组织,石蜡包埋切片,采用免疫组化法检测PDE1A、PDE2A、PDE4A、PDE5A、PDE9A、PDE10A的蛋白表达。结果模型组大鼠海马CA1区的PDE1A、PDE2A、PDE5A、PDE9A蛋白表达显著升高(P<0.05,P<0.01,P<0.01,P<0.01),PDE4A、PDE10A的蛋白表达显著降低(P<0.01,P<0.05),芍药苷可显著下调PDE1A、PDE2A、PDE5A、PDE9A的蛋白表达(P<0.05,P<0.01,P<0.01,P<0.01),明显上调PDE4A、PDE10A的蛋白表达(P<0.05)。结论血虚肝郁证模型大鼠由于受到辐照和束缚双重应激的影响,海马CA1区PDE各亚型表达显著改变,芍药苷能够通过调控不同底物分解酶的表达,从而影响2种核苷酸的含量,表明白芍养血柔肝作用是多靶点多途径的动态平衡。Objective To observe the effects of paeoniflorin on the protein expression of phosphodiesterase( PDE) subtypes in the hippocampus CA1 zone of rats with syndrome of blood deficiency and liver stagnation. Methods The rats were divided into blank group,model group,low dose and high dose of paeoniflorin( low-,high-dose) group. Each group had been intragastrically administered since the first day of modeling—low-dose group with 20 mg / kg paeoniflorin,high-dose group with 40 mg / kg,blank group and model group with pure water,for consecutive 21 days. After perfusion fixation,the brains of all rats were dissected and paraffin-emedded sections of brains were studied. The protein expression of PDE1 A,PDE2A,PDE4 A,PDE5A,PDE9 A and PDE10 A were detected using immunohistochemistrical methods. Results Compared with blank group,the protein expression of PDE1 A,PDE2A,PDE5 A and PDE9 A of the model group increased significantly( P〈0. 05,P〈0. 01,P〈0. 01,P〈0. 01),and that of PDE4 A,PDE10A declined( P〈0. 01,P〈0. 05). Paeoniflorin down-regulated the protein expression of PDE1 A,PDE2A,PDE5 A and PDE9 A remarkably( P〈0. 05,P〈0. 01,P〈0. 01,P〈0. 01),and upregulated that of PDE4 A,PDE10A( P〈0. 05). Conclusion The protein expressions of PDE subtypes of rat model with syndrome of blood deficiency and liver stagnation changed remarkably due to the double stimulations of radiation and restraint. The effect of nourishing blood and liver of White Peony Root( Radix Paeoniae Alba,Baishao) suggested be a dynamic balance of multi-target with multi-path,which was proved by the regulations of paeoniflorin on substrates hydrolases resulted in the concentration of the two nucleotides.
