摘 要:以俄罗斯引进的蓝靛果忍冬优良无性系Berel(L.caerulea var.kamtschatika X altaica)、Rus1(L.caerulea var.kamtschatika)和Rus2(L.caerulea var.regeliana X altaica)为研究对象,以其健壮枝条上的茎尖及茎段为外植体,进行组织培养研究,建立适合俄罗斯蓝靛果忍冬外植体生长、扩繁、生根的基本培养程序.结果表明:俄罗斯蓝靛果忍冬的3个优良无性系以MS+1.0 mg/L 6-BA+0.2 mg/L IBA为初代培养基效果均较好,但继代增殖培养基和生根培养基差异较大.其中,适合Berel增殖的培养基为MS+2.0 mg/L 6-BA+0.2 mg/L IBA,增殖系数达到5.6,生根培养基为WPM+1.5 mg/L IBA;适合Rus1和Rus2继代增殖的培养基为WPM+0.5 mg/L 6-BA+0.2 mg/L IBA,增殖系数达到6.8和7.6,生根培养基为1/2MS+0.2 mg/L IBA;适合俄罗斯蓝靛果忍冬试管苗移栽的基质为草炭土+河沙(体积比1∶1)的混合基质.L.caerulea excellent clone(L.caerulea var.kamtschatika X altaica),Rus1(L.caerulea var.kamtschatika)and Rus2(L.caerulea var.regeliana X altaica) introduced from Russia as the research objects,with its strong branches on the stem tip and stem segments as explants for tissue culture,a complete micro-propagation protocol is developed for Russia L.caerulea.The results showed that the 3 excellent clones of Russian L.caerulea with MS+1.0mg/L 6-BA+0.2 mg/L IBA as primary radical had better effect,but subculture proliferation medium and rooting medium were large differences,which is suitable for training base of the proliferation of Berel MS+2.0 mg/L 6-BA+0.2 mg/L IBA,the proliferation coefficient reached 5.6,rooting culture medium for WPM + 1.5 mg/L IBA and training base for Rus1 and Rus2 proliferation WPM + 0.5 mg/L 6-BA + 0.2 mg/L IBA.The proliferation coefficient reached 6.8 and 7.6,and rooting medium for 1/2MS + 0.2 mg/L IBA,Russia L.caerulea test tube seedling transplanting matrix is mixed matrix of turfy soil + sand(rate of volume 1 ∶ 1).
