摘 要:目的探讨不同剂量氯化甲基汞蓄积时长差异对成年大鼠记忆力及脑组织中Aβ1-42的影响。方法通过灌胃给予大鼠不同剂量的氯化甲基汞,采用水迷宫观察大鼠记忆力变化,采用免疫组织化学方法观察不同时间点各组大鼠脑组织中Aβ1-42的变化。结果氯化甲基汞明显增加学习及记忆潜伏期,并增加了脑组织内β淀粉样蛋白沉积,大剂量组与其他各组比较,差异均有统计学意义(P<0.05,P<0.01),且与汞蓄积量明显相关。结论氯化甲基汞促进Aβ1-42在脑组织中沉积是引起认知损害的机制之一。Objective To investigate the effect of different doses of methyl-mercuric chloride on memory and Aβ1-42 of adult rats in different courses.Methods Adult rats were treated with different doses of methylmercuric chloride by intragastric administration.The changes of memory were observed by water maze.Aβ1-42 was detected by immunohistochemistry at different times after administrating various dosages.Results MMC significantly increased the latency of memory,and increased Aβ1-42,which of large dose group were significantly higher than those of other groups(P〈0.05 or P〈0.01).Conclusions MMC induces Aβ1-42 deposition in brain to cause cognitive impairment,which effects are dose and time dependent.
