金丝桃苷联合长春新碱对人结肠癌HCT8细胞增殖和凋亡的影响

摘　　要：目的研究金丝桃苷联合长春新碱对人结肠癌HCT8细胞增殖和凋亡的影响,探讨其协同增效作用及机制。方法采用噻唑蓝(MTT)法检测不同浓度的金丝桃苷对HCT8细胞的增殖抑制作用,并计算IC20,IC50;采用IC20浓度的金丝桃苷联合不同浓度的长春新碱,观察对HCT8细胞增殖的影响;金丝桃苷联合长春新碱处理HCT8细胞,流式细胞仪检测细胞凋亡率,比色法检测Caspase-3和Caspase-9活性的变化。结果金丝桃苷能有效抑制HCT8细胞的增殖,IC20约为5mg/L,IC50约为30mg/L。金丝桃苷与长春新碱联合作用于HCT8细胞48h,可提高HCT8细胞对长春新碱的敏感性,增敏4.88倍。金丝桃苷与长春新碱联合用药细胞凋亡率、Caspase-3和Caspase-9活性比单独用金丝桃苷和长春新碱都有所提高(P<0.01)。结论金丝桃苷能提高人结肠癌HCT8细胞对长春新碱的敏感性,金丝桃苷联合长春新碱后会增强Caspase-3和Caspase-9活性,从而促进HCT8细胞凋亡。Objective To investigate the synergistic effect of hyperin combined with vincristine on proliferation and apoptosis in human colon cancer HCT8 cell line and the possible mechanism. Methods MTT assay was used to measure the proliferation inhibition effects of hyperin and hyperin combined with vincristine on HCT8 cells. Apoptotic population was evaluated by flow cytometry and the changes of Caspase- 3 and Caspase- 9 activities were detected by colorimetric assays. Results IC20 and IC50for hyperin against HCT8 cells were 5mg / L and 30 mg / L,respectively. The chemo- therapeutic effects of vincristine on HCT8 cells were enhanced when combined with hyperin,and the drug sensitivity was increased by about 4. 88 times. The apoptotic population and Caspase- 3 and Caspase- 9 activities in combination groups were increased compared to vincristine group or hyperin group（ P〈0. 01）. Conclusion Hyperin has synergistic inhibitory effect with vincristine on HCT8 cells. Hyperin combined with vincristine might increase Caspase- 3 and Caspase- 9 activities and induce cell apoptosis.
