摘  要:目的研究兔同种异体骨经多聚赖氨酸表面改性后,对兔骨髓间充质干细胞(BMSCs)亲和性的变化。方法采取冻干法,将多聚赖氨酸覆盖在同种异体骨的微孔表面,对其进行表面改性。表面改性后的同种异体骨设定在实验组,表面未改性的设定在对照组,BMSCs分别种植于两组同种异体骨的微孔表面。采用扫描电镜观察、噻唑蓝(MTT)法检测BMSCs在同种异体骨微孔表面贴附、增殖情况,同时检测两组经诱导培养后的细胞表达的碱性磷酸酶(ALP)的活性,并对两组的结果进行比较。结果实验组中BMSCs在同种异体骨微孔表面贴附、增殖以及向成骨细胞分化的情况明显优于对照组(P<0.05)。结论同种异体骨经多聚赖氨酸表面改性后,显著提升了BMSCs的亲和性。Objective To investigate the change of the affinity of allogeneic bone to BMSCs of the rabbit after surface modification with polylysine. Methods The surface modification of the allogeneie bone was processed by the polylysine with lyophilization methods. The allogeneic bones whose surface was modifieated were taken as the experimental group, and the others without modification were taken as the control group. The BMSCs was implanted onto the surface of the micropores in the bone allografts. The attachment and proliferation of BMSCs on the surface of the mieropores in the bone allografts were observed with SEM and detected with MTT methods, and the activity of the ALP expressed by the osteoblast - induced BMSCs was also detected. Then, the results were compared. Results The attachment, proliferation and osteoblast-differentiation of BMSCs in the experimental group were much superior than those in the control group( P 〈0.05 }. Conclusion The affinity of the bone allografts to BMSCs can be significantly promoted after surface modification with polylysine.
